摘要
目的:观察阳和化岩汤对乳腺癌大鼠Fas、FasL和Bcl-2表达的影响。方法:应用7,12-二甲基苯并蒽(DMBA)诱导大鼠建立乳腺癌模型,药物干预4周后,HE染色观察大鼠乳腺组织形态学变化,免疫组化法检测Fas、FasL、Bcl-2蛋白表达的阳性率,Q-PCR法检测Fas、Bcl-2 mRNA表达。结果:阳和化岩汤可明显降低大鼠乳腺癌的发生率(P<0.05),同时可升高Fas的表达(P<0.05)、降低FasL和Bcl-2的表达(P<0.05)。结论:阳和化岩汤具有预防乳腺癌的作用,其作用机制可能与调控细胞凋亡相关蛋白Fas、FasL和Bcl-2表达相关。
Objective:To observe the effect of Yanghe Huayan decoction on the expression of Fas/FasL and Bcl-2 in breast cancer model rats. Methods:Breast cancer rat models were established by using 7,12-dimethylbenzanthracene(DMBA),and then the rats were treated for 4 weeks. The rat breast tissue was observed after HE staining for morphological changes,and the positive rate of Fas/FasL and Bcl-2 expression were detected with immunohistochemistry,and expressions of Fasand Bcl-2 m RNA were detected with quantitative PCR. Results:Yanghe Huayan decoction could reduce the incidence rate of breast cancer in rats(P〈0.05),increase the expression of Fassignificantly(P〈0.05) and decrease expressions of FasL and Bcl-2 significantly(P〈0.05). Conclusion:Yanghe Huayan decoction could prevent breast cancer,and the mechanism might be involving regulating apoptotic related proteins Fas/FasL and Bcl-2.
