左、右归丸及其拆方对去卵巢大鼠肾脏TGFβ1/Smad1表达的影响

Effect of Zuogui Pill,Yougui Pill and Their Disassembled Prescriptions on Kidney Index in Ovariectomy Rats

目的:观察左、右归丸及其拆方对去卵巢大鼠肾脏TGFβ1/Smad1基因与蛋白表达的影响,并探讨补肾法对去卵巢模型大鼠肾脏TGFβ1/Smad1通路的影响。方法:将90只SD雌性大鼠随机分为空白组、假手术组、模型组、补佳乐组、左归丸组、右归丸组、共同药组、滋阴药组、补阳药组。除假手术、空白组外均进行双侧卵巢切除。经12周治疗分别以Q-PCR法、Western blot法检测TGFβ1、TβRⅠ、TβRⅡ、Smad1在肾脏中基因和蛋白的表达。结果:与空白组相比,除假手术组外,TGFβ1、TβRⅠ、TβRⅡ、Smad1 mRNA及蛋白表达降低(P<0.01);与模型组比较,各用药组TGFβ1、TβRⅠ、TβRⅡ、Smad1 mRNA及蛋白表达升高(P<0.01);与补佳乐组比较,各用药组TGFβ1、TβRⅠ、TβRⅡ、Smad1表达低,其中左归丸组表达较高。结论:左、右归丸及其拆方可影响肾脏中TGFβ1/Smad1基因与蛋白的表达,其中以左归丸改变最为显著。

Objective ：To observe Zuogui Pill, Yougui Pill and their disassembled prescriptions on osteoporosis in ovariecto- mized rat kidney and explore methods of prevention and treatment of osteoporosis and kidney disease. Methods ： Ninety SD female rats were randomly divided to 9 groups ： Control group, Sham group, ovariectomy group, BJL group, ZGP group, YGP group, GTY group,ZYY group, BYY group, 10 rats in each group. Except Control group and Sham group, other groups established ovariecto- mized models. After 12 weeks,use real -time quantitative PCR and Western blot to detect the gene and protein expressions of TGFI31, TβR I , TβR Ⅱ and Smadl in the kidney. Results：Compared with Control group, the gene and protein expressions of TGFβ1 ,TIER I ,TβR Ⅱ and Smadl of OVX group were lower （P 〈0. 01 ）. Compared with OVX group,the gene and protein ex- pressions of TGFβ1, TβR I , TβR Ⅱ and Smadl of each treatment group were improved （ P 〈 0. 01 ） and those of ZGW group were better. Conclusion：Zuogui Pill, Yougui Pill and their disassembled prescriptions can influence the treatment of kidney TGFβ1/ Smadl pathway expression and Zuogui Pill is more notable.
