摘要
为了对麝属动物源性成分进行有效鉴定,根据麝属动物线粒体DNA(mt DNA)序列,使用分子生物学软件Oligo7.0设计了特异性引物,用来建立PCR检测麝属动物源性成分的方法。结果显示:使用麝特异性引物优化的PCR体系可以对两份麝样品进行扩增,凝胶电泳均出现特异性条带。方法特异性好,只扩增麝属动物样品,16份其他物种的样品均为阴性结果。方法灵敏度较高,可检测到100个拷贝DNA的数量级。表明本研究建立的方法可应用于动物组织及其加工产品中的麝属动物源性成分的检测。
In order to effectively identify Moschus-origined ingredients, the specific primers were de- signed using molecular biology software Oligo7.0 based on mitochondrial DNA sequence of Moschus. PCR assay for detecting Moschus-origined ingredients was established with newly designed and optimized primers. Two samples of Moschus were amplified using the primers, and the specific bands appeared in gel electrophoresis. The assay had high specificity, for it could amplify samples only con- taining Moschus-origined ingredients, all the other sixteen samples were negative. Furthermore, the assay had high sensitivity, for the minimum detectable amount of DNA was 100 copies DNA template. The results indicated that this assay could be applied to detect Moschus-derived ingredients in animal tissues and related processed products.
