Bromothymol Blue Decolorization Rates and Evaluation of Mycelial Biomass Production,Flavone Levels and Antioxidant Activity of Ten Sanghuangporus Strains

摘要 采用色度培养基(liquid medium supplemented with bromothymol blue and lactose,LBL)对10株桑黄菌株进行脱色能力评价及可见光谱分析,探究了菌株LBL法脱色率与液体发酵菌丝体生物量、菌丝体黄酮含量及菌丝体醇提物清除自由基活性的相关性。结果表明:脱色率较低的菌株,在液体传代发酵过程中生物量呈下降的趋势;脱色率与菌丝体生物量(第一代)、菌丝体黄酮含量、醇提物清除超氧阴离子IC_(50)值、醇提物清除过氧化氢IC_(50)值相关系数分别为0.44,0.73,-0.83和-0.55。数据说明可以通过LBL法快速评价桑黄菌株活性并筛选出衰退菌株。 Bromothymol blue-decolorizing ability of metabolites produced during the growth of ten Sanghuangporus strains in a liquid culture medium(20% potato,2% NH4NO3,0.05% MgSO4·7H2O,0.15%KH2PO4,pH 7.0)supplemented with the 0.006% dye and 2% lactose(LBL medium),together with mycelial biomass production,mycelial flavone levels,and free radical scavenging activity of ethanolic extracts of fungal mycelia,were determined.Mycelial biomass production decreased with increasing frequency of subculture for strains with low dye-decolorizing ability,and the correlation coefficients between decolorizing rate and mycelium biomass,mycelial flavone levels,and IC50 values for superoxide anion radical and H2O2 radical scavenging activity of ethanolic extracts of fungal mycelia,were 0.44,0.73,-0.83and-0.55,respectively.Our data revealed that dye-decolorization rates in LBL medium can be used to evaluate these biological features in strains of Sanghuangporus.
