摘要
目的探讨电针刺激对糖尿病神经病理性疼痛大鼠痛阈的影响。方法 128只雄性SD大鼠随机均分为4组(n=32):糖尿病神经病理性疼痛组(D组)造模成功后不给予治疗;电针刺激组(EA组)造模成功后给予电针治疗;假电针刺激组(A组)只将针灸针插入穴位,但不给予电刺激;假穴位电针刺激组(E组)将电针插入穴位右侧1 cm处,并给予电刺激,记录电针干涉前(T1)、电针干涉7 d(T2)、14 d(T3)及21 d(T4)大鼠的机械缩足反射阈值及血糖的水平;各时间点测定结束后,每组随机选取8只大鼠取L4-6脊髓,用Western blot的方法测定脊髓p38 MAPK表达水平,分析痛阈变化及p38 MAPK表达水平的相关性。结果与D、A、E组相比,EA组T2、T3、T4时间点痛阈均较高,差异有统计学意义(P<0.05);与D、A、E组相比,EA组T2、T3、T4时间点p38 MAPK活化水平均下降,差异有统计学意义(P<0.05);DNP大鼠痛阈及p38 MAPK变化呈负相关(r=-0.942,P<0.05)。结论电针刺激能提高糖尿病神经病理性疼痛大鼠的痛阈,其机制可能与抑制脊髓p38MAPK的表达有关。
Objective To investigate the effects of electroacupuncture stimulation on analgesic for diabetic neuropathic pain in rats. Methods A total of 128 male adult SD rats were randomly divided into 4 groups (n = 32) : diabetic neuropathic pain model group (Group D) and electroacupuncture stimulation group (Group EA), sham EA stimulation group (Group E), and sham acupuncture point group (Group A). The changes of mechanical shrinkage threshold, weight and blood glucose on the day before acupuncture stimulation and 7 days, 14 days, 21 days after electroacupuncture stimulation were recorded. Eight random rats from each group were sacrificed for L4 -6 spinal cord to inspect p38MAPK expression levels in the spinal cord. The correlation between pain threshold and p38 MAPK expression were analyzed. Resuits Compared with Group D, E and A, significantly higher point thresholds were observed in Group EA at T2, T3, and T4 (P 〈 0.05 ). Compared with Group D, E and A, significantly p38 MAPK activation levels were observed in Group EA at T2, 33 and T4 (P 〈 0.05 ). Pearson correlation coefficient for pain threshold and p38 MAPK was - 0. 942. Conclusion Electroacupuncture can improve the pain threshold in DNP rats, the mechanism may be related to the inhibition of microglia p38MAPK expression in spinal cord.
