摘要 目的研究羟基红花黄色素A(HSYA)对人脑胶质瘤U251细胞增殖、迁移和侵袭的影响,并探讨其作用机制。方法体外培养人脑胶质瘤U251细胞,经不同浓度(25、100、400μmol/L)HSYA处理后分别采用四甲基偶氮唑蓝法(MTT)、划痕试验和Transwell试验观察HSYA对U251细胞增殖、迁移和侵袭的影响,采用Western blot方法检测基质金属蛋白酶(MMP)-2、MMP-9的蛋白表达水平。结果与空白对照组比较,HSYA能降低U251细胞存活率,缩短细胞迁移距离,减少U251细胞穿膜数,差异均有统计学意义(P<0.01或P<0.05),同时,HSYA能下调MMP-2、MMP-9的蛋白表达。结论 HSYA对脑胶质瘤U251细胞增殖、迁移及侵袭等具有抑制作用,其作用机制可能与下调MMP-2、MMP-9表达有关。 Objective To investigate the effects of hydroxysafflor yellow A (HSYA) on proliferation, migration and inva- sion of glioma U251 cells, and to explore its mechanism. Methods The human glioma U251 cells were cultured in vitro and treated with different concentrations of HSYA (25, 100, 400 μmol/L), then the methyl thiazolyl tetrazolium (MTF) assay, scratch test and Transwell assay were used to observe the effects of HSYA on proliferation, migration and invasion of U251 cells respectively, and the expression levels of matrix metalloproteinase (MMP)-2 and MMP-9 were ex- amined by Western blot assay. Results Compared with blank control group, the U251 cell viability in HSYA group was decreased, the migration distance was shortened, the number of penetrated cells was reduced obviously, the differences were all statistically significant (P 〈 0.01 or P 〈 0.05). Moreover, HSYA could decrease the expression of MMP-2 and MMP-9. Conclusion HSYA has inhibitory effects for the proliferation, migration and invasion of glioma ceils, and the mechanism may be related to the inhibition of MMP-2 and MMP-9 expression.
