金雀异黄素调节围绝经期小鼠血清抑制素与性激素水平的研究 被引量:3

Studies on the Regulating Effect of Genistein on Inhibin and Sex Hormones in Perimenopausal Model Mice

摘要 目的:研究金雀异黄素对围绝经期模型小鼠血清抑制素与性激素水平的调节作用。方法:选择12月龄自然衰老ICR小鼠作为围绝经期动物模型,分为对照组(CG)、Gen低(L-Gen)、中(M-Gen)、高剂量组(H-Gen)及雌激素组(EG)。剂量组分别灌胃给予金雀异黄素(Gen)15,30,60 mg/kg,雌激素组灌胃给予戊酸雌二醇0.5 mg/kg,持续8周。采用酶联免疫吸附法(ELISA)检测各组小鼠血清中抑制素A(Inh-A)、抑制素B(Inh-B)以及性激素水平;免疫组织化学法检测小鼠卵巢组织中FSHR和LHR的表达情况。结果:与对照组相比,金雀异黄素能提高小鼠血清中Inh-A和Inh-B的水平;提高雌二醇(E2)和孕酮(P)水平,降低睾酮(T)、促卵泡成熟激素(FSH)和促黄体生成素(LH)水平;增强促黄体生成素受体(FSHR)表达和降低促卵泡成熟激素受体(LHR)的表达,其作用效果与雌激素相似。结论:在金雀异黄素试验剂量条件下,对围绝经期小鼠血清中抑制素和性激素水平均有调节作用,并影响小鼠卵巢组织中相应受体的表达,从而调节围绝经期小鼠卵巢功能。 Objective: The regulating effect of genistein(Gen) on inhibin and sex hormones in perimenopausal model mice was studied in this paper. Methods: Twelve months natural aging ICR mice was selected as the menopausal animal models. They were divided into control group(CG), low dose Gen group(L-Gen), middle dose Gen group(M-Gen),high dose Gen group(H-Gen) and estrogen group(EG). Mice in dose group received genistein at 15, 30, 60 mg/kg respectively by gastric gavage and mice in EG received pentanoic acid estradiol 0.5 mg/kg for 8 weeks. Concentration of Inh-A, Inh-B and sex hormones were detected by enzyme-linked immunosorbent(ELISA). FSHR protein and LHR protein of ovary were examined by immunohistochemical method. Results: Compared with control group, the levels of estradiol and progesterone raise and the levels of testosterone declined in Gen group, there is significant difference(p〈0.05),the expression of FSHR protein in mice' ovarian increased, LHR protein expression decreased, which was similar to estrogen. Conclusions: Under the current dose, Gen has regulating effect on inhibin and sex hormones of menopausal mice and also had effect on FSHR protein and LHR protein in mice' ovarian tissue, which considered to be the cause of adjusting the perimenopausal ovarian function in mice.
