辛伐他汀对大鼠蛛网膜下腔出血后脑血管痉挛的干预作用研究 被引量:3

摘要 目的探究辛伐他汀对大鼠蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)的干预作用。方法选取健康3月龄SD雄性大鼠30只,随机分为A组(辛伐他汀组)、B组(SAH组)、C组(假手术组),各10只。入选大鼠均于注射第7天后灌注处死,以TUNEL(原位末端标记法)法检测BA平滑肌细胞的凋亡率;取基底动脉行HE(苏木精-伊红)染色,测量其管壁厚度和内径周长。结果与C组相比,A、B两组的管壁厚度及平滑肌细胞凋亡率均增加、内径周长减少,且B组管壁厚度增加量及内径周长减少量明显大于A组、平滑肌细胞凋亡率明显低于A组,差异均有统计学意义(P<0.05)。结论辛伐他汀可抑制平滑肌细胞增殖,减少SAH-CVS的发生,可对大鼠蛛网膜下腔出血后脑血管痉挛起到一定的缓解作用。 Objective To explore intervention effect of simvastatin on cerebral angiospasm after subarachnoid hemorrhage of rat. Methods Selected three months old healthy male SD rats as the object, and all the 30 rats were divided into A group (simvastatin group), B group (SAH group), C group (sham operation group), 10 rats in each group. The rats were injected to death on the seventh day, and apoptosis rate of BA smooth muscle cell was detected by TUNEL. Basilar artery was stained by HE (hematoxylin-eosin) and the thickness of pipe and the inner circumference of the basilar artery were measured. Results Compared with C group, the thickness of pipe and apoptosis rate of A, B groups increased and the inner circumference decreased. The increment of the thickness of pipe and the decrement of the inner circumference of the hasilar artery of the B group were greater than those of A group, and apoptosis rate of B group was lower than that of A group (P〈0.05). Conclusion Simvastatin can inhibit smooth muscle cell proliferation, decrease the occurence of SAH-CVS, and ease cerebral angiospasm after subarachnoid hemorrhage of rat.
