静电场对钝顶螺旋藻藻蓝蛋白稳定性影响研究

摘要 为了研究静电场对钝顶螺旋藻藻蓝蛋白稳定性的影响,通过静电场对钝顶螺旋藻进行诱变处理,处理条件为5 min(0、3、6、9、12、15、18、21、24、27 k V)的藻珠IS、ES-1、ES-2、ES-3、ES-4、ES-5、ES-6、ES-7、ES-8、ES-9,从电场处理的藻种中选取了一株优质藻珠ES-8,与出发株IS比较:其β-胡萝卜素含量增幅达到32.41%;藻蓝蛋白含量增幅达到19.26%;藻蓝蛋白紫外吸收光谱的最大吸收峰位置一致,最大吸收峰值略高于IS。在藻蓝蛋白的稳定性试验中,ES-8与IS在p H 4~10范围内其藻蓝蛋白都能保持较高的活性;ES-8与IS在低于25%的乙醇溶液中,其藻蓝蛋白具有较好的稳定性,保存率分别在84.99%与82.08%以上;在浓度低于4%的氯化钠溶液中,IS株的藻蓝蛋白的稳定性变化不大,ES-8株的藻蓝蛋白受到非常好的保护。 Influences of electrostatic field on characteristics of phycocyanin of Spirulina platensis were studied by mutagenizing Spirulina platensis for 5 min under the electrostatic field conditions:0,3,6,9,12,15,18,21,24 and 27 k V,respectively,then the algal strains called IS,ES-1,ES-2,ES-3,ES-4,ES-5,ES-6,ES-7,ES-8 and ES-9 were obtained;among others,ES-8 A which was deemed high quality was selected to compare with the starting strain IS.The findings suggested that the content of β-carotene of ES-9 increased by 32.41%,content phycocyanin increased by 19.26%;and the maximum absorption peak position of ultraviolet absorption spectrum of the phycocyanin kept the same while the maximum absorption peak was slightly higher than that of IS.With regards to the test of phycocyanin stability within the range of p H 4 ~10,both ES-8 and IS maintained high activity;on the other hand,in the ethanol solution with a concentration of less than 25%,both ES-8 and IS had good stability with the preservation rate of over 84.99% and 82.08%,respectively;while in sodium chloride solution with concentrations less than 4%,changes of stability were scarcely observed in IS strain,and ES-8 strain was well protected.
