枳椇水提液不同极性萃取组分对乙醇诱导的人体HL7702肝细胞脂肪变性的影响

摘要 探讨枳椇水提液不同极性萃取组分对乙醇诱导的人体HL7702肝细胞脂肪变性的影响。体外培养人正常肝细胞株HL7702,设立空白对照组、乙醇模型组和枳椇不同极性萃取液干预组,采用MTT法测定细胞存活率,油红O染色观察细胞内脂滴形成情况,检查细胞内TG、ALT和AST的含量。结果 0.6%乙醇诱导HL7702细胞24 h成功建立脂肪变性模型。同空白对照组相比,乙醇模型组细胞内TG、ALT和AST含量明显增高,差异有统计学意义(P<0.05);同模型组比,枳椇乙酸乙酯层萃取液干预的细胞TG、ALT和AST含量明显降低,差异有统计学意义(P<0.05)。结论:具有减轻乙醇细胞损伤和降低肝细胞脂肪变性作用的最佳组分是乙酸乙酯萃取层。 To determine the effects of polar extraction layer of Hovenia acerba at different concentrations on alcohol-inducedlipogenesis in HL7702 liver cells.Cultured HL7702 liver cells were divided into eleven groups:blank control group,alcohol induc-tion group,and polar extraction layer of Hovenia acerba intervention groups.Cell proliferation was measured by MTT assay.The ac-cumulation of lipid droplets was observed by light inverted microscopy after red oil-O staining. The levels of TG,ALT and ASTwere determined using ELISA kits.A model of alcohol-induced steatosis was successfully induced in HL7702 liver cells by incuba-tion with 0.6% ethanol for 24 h.Compared with normal group,ALT,AST and TG were increased in alcohol induction group,and dif-ference was statistically significant(P〈0.05). Compared with alcohol induction group,the liquid Hovenia dulcis and ethyl acetateextract layer intervention ALT、AST and TG were significant decreased(P〈0.05).The Ethyl acetate extraction layer can relieve eth-anol cell damage and reduce the hepatocyte fatty degeneration in HL7702 fat cell induced by ethanol.
