摘要
目的研究脱氧胆酸(DCA)对法尼酯衍生物X受体(FXR)的作用及其对结肠癌细胞生物学行为的影响。方法分别采用噻唑兰比色分析(MTT)法和蛋白质印迹法(Western Blot)检测不同浓度DCA对人结肠癌上皮细胞(Caco-2)增值率和FXR、尾侧型同源转录因子2(Cdx2)及黏蛋白2信使RNA(MUC2m RNA)表达水平的影响,并分析其对结肠癌细胞生物学行为的影响。结果DCA浓度10 umol/L时在6 h、12 h、24 h、48 h及72 h不同时间点Caco-2细胞抑制率均显著低于其他浓度组,差异有统计学意义(P<0.05)。同浓度下72 h时Caco-2细胞抑制率显著高于其他各时间点,差异有统计学意义(P<0.05)。72 h时10 umol/L组FXR为(2.02±0.33)%,显著低于其他浓度组,Cdx2及MUC2表达水平分别(3.97±0.67)%及(3.76±0.48)%,均显著高于其他浓度组,差异有统计学意义(P<0.05)。结论 DCA能促进Caco-2细胞增殖,DCA影响结肠癌患者细胞增殖率可能是通过影响FXR及其对Cdx2等因子表达水平来实现的。
Objective To study effects of deoxycholic acid(DCA) on farnesoid x receptor(FXR) and the biological behavior of colon cancer cells. Methods Using the MTT colorimetric analysis(MTT) method and western blot to detect effects of different concentrations of DCA on appreciation rate of colon cancer epithelial cells(Caco-2), FXR, caudal type homologous transcription factor 2(Cdx2) and mucin 2 messenger RNA(MUC2m RNA)expression level. And its effects on the biological behavior of colon cancer cells were analyzed. Results At DCA concentration of 10 umol / L, appreciation rate of Caco-2 at different time points(6 H, 12 h, 24 h, 48 h and 72h) were significantly higher than other concentration groups( P〈0.05). After 6h, appreciation rate of Caco-2 at the same concentration was significantly higher than other time points( P〈0.05). After 6 hours, expression level of FXR, Cdx2 and MUC2 were(10.45 ±3.27)%,(12.34 ±2.31)% and(13.06 ±2.24)% in 400 umol / L group, which were significantly higher than other concentration groups( P〈0.05). Conclusion Short time, low concentration of DCA can promote the proliferation of Caco-2 cells, long time, high concentration of DCA can inhibit the proliferation of Caco-2 cells. Effects of DCA on appreciation rate of patients with colon cancer may be achieved through the impact of expression level of FXR, Cdx2 and other factors.
