摘要
为了研究含黄素单氧化酶3(FMO3)基因T329S突变位点在河北太行鸡不同群体中的等位基因及基因型频率分布情况,采用聚合酶链式反应-限制性片段长度多态性分析方法(PCRRFLP)和DNA测序技术分别对赞皇、平山、唐县地区3个河北太行鸡群体FMO3基因及基因型频率进行检测。结果显示,平山地区太行鸡群体中未检测到SS型个体,而平山、唐县地区太行鸡群体中均有FMO3基因SS型个体;赞皇、平山、唐县3个太行鸡群体中T等位基因的频率分别为0.129 8、0.075 0、0.084 2,其频率均远低于等位基因A的频率。独立性卡方检验结果显示,不同群体间基因型频率分布差异不显著;经适合性卡方检验,赞皇、平山、唐县3个太行鸡群体卡方值分别为1.173 0(P>0.05)、0.035 1(P>0.05)、0.188 5(P>0.05),说明该基因位点A→T碱基突变在3个群体中均处于Hardy-Weinberg平衡状态;通过分析3个不同群体遗传变异参数发现,3个太行鸡群体在该基因位点上均处于低度多态(PIC<0.25)。以上结果表明,河北太行鸡群体中FMO3基因型频率较低,可以通过PCR-RFLP的方法予以剔除。
In order to study the distribution of T329S mutation in flavin-containing monooxygenase 3 ( FM03) in different populations of Taihang Chickens in Hebei. Polymerase chain reaction -restriction fragment length polymorphism ( PCR-RFLP) followed by DNA sequencing assay was used for detecting the frequency of genotype and allele of FM03 gene in three populations of Taihang Chickens. The results showed that no individual would lay fishy taint eggs in Taihang Chickens of Pingshan, fishy taint suscepti-ble individuals with genotype SS were detected in another two groups. The frequency of T allele was 0. 129 8, 0. 075 0 and 0. 084 2, which was much lower than A allele, respectively. The Chi-square test result showed that there were no significant differences among the three populations with its genotype frequencies ,and the Chisquare value were 1.173 0(P〉0. 05),0. 035 1(P〉0.05) and 0 .1 8 8 5(P〉0.05), which were fit with Hardy-Weinberg equilibrium in different populations of Taihang Chickens in Hebei. As for the genetic parameters showed that the three populations of Taihang Chickens were all intermediate polymorphism on this locus. In conclusion, the frequencies of fishy taint genotype of Taihang Chickens in Hebei was not high and the susceptible individuals could be easily detected by PCR-RFLP method.
