沙棘种子萌发的基本培养基的优化 被引量:4

摘要 以沙棘"实优一号"品种的种子作为外植体,经过前期的消毒处理后分别接种在MS、1/2 MS、1/4MS培养基中,在11℃培养10 d后移入25℃中培养,接种培养30 d后观察统计种子在不同基本培养基中的萌发率、萌发时间以及胚根生长状况,从而确定沙棘种子萌发的最佳培养基类型。结果显示:在相同培养条件下,1/2 MS培养基中的沙棘种子在培养7 d后最先萌发,培养30 d后其萌发率最高(16.67%),胚根分化出2-4个分支且生长状况最佳;1/4 MS培养基次之;MS培养基最差。因此,1/2 MS培养基为沙棘"实优一号"品种种子萌发的最佳基础培养基。 The seeds of "Shiyou No.1"sea buckthorn(Hippophae rhamnoides) cultivar,which are taken as explants,were inoculated respectively in MS,1 /2 MS,1 /4 MS medium after the previous sterilization,and cultivated in 11 ℃ for ten days and then 25 ℃ for 20 days.In order to determine the optimal medium in which sea buckthorn seeds germinated after cultivating for 30 days,the germination rates and time of the seeds,and the condition of radicle growth in different medium were observed and counted.The results showed that under the same culture condition,the seeds firstly germinated in the 1 /2 MS medium after cultivating for seven days,and the germination rate of the seeds was highest(16.67 %) after cultivating for 30 days.Moreover,in1 /2 MS medium,the radicles differentiated 2 ~ 4 branches whose growth conditions were the best.However,the seeds in 1 /4 MS medium took the second place,and that in MS medium were the worst.Thus,the 1 /2MS medium is the best basic medium for seeds germination in"Shiyou No.1"sea buckthorn cultivar.
