摘要
目的观察可降解涂层材料聚乙交酯-丙交酯(PLGA)和聚三亚甲基碳酸酯(PTMC)对人血管内皮细胞和平滑肌细胞增殖行为的影响。方法在316L不锈钢试样表面制作PLGA和PTMC涂层薄膜,同时设立316L不锈钢试样对照组,分离人脐静脉内皮细胞和脐动脉平滑肌细胞,和上述不同试样共培养,CCK-8检测共培养1d和3d后各组内皮细胞和平滑肌细胞吸光度值;分别对内皮细胞和平滑肌细胞进行免疫荧光染色,荧光显微镜观察共培养1d和3d后不同样品表面细胞形貌差异,从而评价不同涂层材料对内皮细胞和平滑肌细胞增殖行为的影响。结果和对照组相比,在不同时间点,PLGA和PTMC涂层表面内皮细胞和平滑肌细胞CCK-8吸光度值均明显降低,表面细胞数量均有明显减少,差异有统计学意义(P<0.01);和PLGA涂层表面相比,PTMC涂层表面内皮细胞和平滑肌细胞CCK-8吸光度值更低,增殖的细胞更少(P<0.01)。结论 PLGA和PTMC涂层表面均不利于内皮细胞和平滑肌细胞生长,尤其以PTMC涂层表面更为明显。
Objective To observe the influence of biodegradable coating materialspolyglycolide-lactide( PLGA) and polytrimethylene carbonate( PTMC) on the proliferation behavior of human vascular endothelial cells and smooth muscle cells. Methods PTMC and PLGA coating film were produced on the surface of 316L stainless steel,and 316L stainless steel samples were set up as the control group. Human umbilical vein endothelial cells and human umbilical artery smooth muscle cells were separated and cultured with different samples. Then,detecting absorbance values of each group of endothelial cells and smooth muscle cells with CCK-8 1d and 3d after culturing. The morphological dfferencens of staining endothelial cells and smooth muscle cells were respectively by observed fluorescence microscopy on different sample surfaces 1d and 3d after,after mcubation for. Results Compared with control group,at different time points,CCK-8 absorbance values of endothelial cells and smooth muscle cells on the coating surface of PLGA and PTMC hwere significantly lower and the number of cells on the coating surface both decreased significantly(P〈0. 01). Compared with coating surface of PLGA,CCK-8 absorbance values of endothelial cells and smooth muscle cells on the coating surface of PTMC hwere lower,which also had less cell proliferation(P〈0. 01). Conclusion PLGA and PTMC coating surface both were not conducive for the growth of endothelial cells and smooth muscle cells,particularly,the PTMC coating was more obvious.
