摘要
目的:初步探讨Survivin、CTNNB1、h TERT表达与胃癌药物敏感度的关系。方法:采用三维立体组织培养药物反应性试验检测20例胃癌组织多西紫杉醇、异长春花碱、伊立替康和奥沙利铂四种药物的敏感度,RT-PCR方法检测胃癌组织Survivin、CTNNB1和h TERT mRNA表达水平。结果:异长春花碱、伊立替康和奥沙利铂三种药物最佳检测浓度分别为20μg/ml、80μg/ml和40μg/ml。CTNNB1 mRNA表达用于判断多西紫杉醇和异长春花碱敏感度的准确率为70%、Survivin mRNA表达用于判断奥沙利铂敏感度的准确率为70%。结论:CTNNB1表达水平和多西紫杉醇与异长春花碱敏感度相关,Survivin表达水平与奥沙利铂敏感度有关。
Objective To investigate the sensitivity of gastric cancer to docetaxel, vinorebine, irinotecan and oxali- platin,and then to assess the relationship between Survivin,CTNNB1 ,hTERT mRNA expression and the chemosensi-tivity. Methods : Histoculture drug response assay was used to assess the chemosensitivity of 20 gastric cancer samples to docetaxel, vinorebine, irinotecan and oxaliplatin. Real - time reverse transcription polymerase chain reaction was used to determine the expression levels of Survivin,hTERT and CTNNB1. Results:The best test concentrations of vi-norebine, irinotecan and oxaliplatin for HDRA were 20,80 and 40|jLg /ml, repectively. The accuracy for CTNNB1 to predict the sensitivity to docetaxel and vinorebine was 7 0 % and the accuracy for Survivin to predict the sensitivity to oxaliplatin was 70%. Conclusion: CTNNB1 may afford to predict the chemosensitivity to docetaxel and vinorebine, while Survivin may predict the chemosensitivity to oxaliplatin in gastric cancer.
