摘要
为了解滇池蓝细菌生物多样性,于2010年6月从滇池采集水样,通过平板分离纯化及液体培养得到一株蓝细菌。该菌在液体培养下呈聚集状态,显微镜镜检观察显示该菌呈丝状,并发绿色及橙红色荧光。以该菌总DNA为模板,用细菌16S rDNA通用引物进行扩增,得到长度约1.5 Kb的序列,将该序列连接到pMD 18-T载体后测序。测序结果与Gen Bank比对显示该菌属于颤藻目细鞘丝藻属的一个新种。
The water samples were collected from Dianchi Lake in June of 2010 for the purpose of investigating the biodiversity of cyanobacteria in this ecosystem. The samples were inoculated in BG-11 medium in artificial light incubator. One cyanobacterial strain was obtained from the water samples. The isolated strain was filamental in morphology observed under microscopy. It had green and red fluorescence when observed by fluorescence microscopy. About 1.5 Kb length of 16 S rDNA sequence was obtained by PCR with bacterial universal primer. The DNA product was introduced into p MD 18-T vector and sequenced. The 16 S rDNA sequence was clustered in Genebank. The phylogenic tree was constructed. Our result indicated that the cyanobacterium DC0129 was a new species that belongs to Oscillatoriales leptolyngbya.
