摘要
以火草幼芽为外植体,对其进行组织培养,以MS为基本培养基,设置不同消毒时间处理,获得了无菌外植体。采用不同激素浓度配比,筛选出各个时段适合的培养基配方:初代培养基为MS+6-BA0.4 mg/L+NAA0.2mg/L,诱导率为55%;增殖培养基为MS+6-BA0.6mg/L+NAA0.1mg/L,增殖系数为3.68;生根培养基为MS+NAA0.3 mg/L,生根率为90%。生根苗炼苗移入红土+腐殖土(1∶1)基质中,移栽成活率达95%以上。
This study conducted a tissue culture test by taking fire weed shoots as explants, MS as medi- um, and setting a different time disinfection treatment to obtain aseptic explants. By using different hor- mone concentration screened, we obtained various periods media formulations: the first generation medi- um was MS + 6-BA0.4 mg/L + NAA0.2rag/L, of 55% inductivity; proliferation medium was MS + 6- BA0.6mg/L + NAA0. 1mg/L, the proliferation coefficient was 3.68 ; rooting medium was MS + NAA0.3 mg/L, of 90% the rooting rate. The results also showed that survival rate of rooting seedling was more than 95% while in the matrix of hardening clay + humus ( 1/1 ).
