摘要
以浙东白鹅为研究对象,在木瓜蛋白酶和风味蛋白酶复合一次酶解鹅皮胶原蛋白基础上,选取4种常见蛋白酶对胶原蛋白酶解产物进行二次酶解,并优化二次酶解条件以提高其产物的抗氧化活性,以抗氧化能力为指标确定二次酶解中的最佳水解酶.在单因素试验基础上,以还原力为指标,选取p H、酶解温度和酶解时间为考察因素,利用响应曲面法确定最佳酶解条件.结果表明,在胶原蛋白的二次酶解中,以碱性蛋白酶酶解所得产物抗氧化活性较好,响应曲面法确定碱性蛋白酶最优酶解条件为p H 8.96、酶解温度49.3℃、酶解时间5.7 h,此时所得多肽还原力达到13.48×10-2,较优化前提高了3.51%.
In this study, on the basis of enzymolysis with papain and flavourzyme of collagen from the skin of Zhedong white goose, four common hydrolasis solutions were used for two-step enzymolysis. Subsequently the conditions of two-step enzymolysis were optimized and the suitable hydrolyase was selected by antioxidant ability. By employing a single-factor test, pH, reaction temperature and reaction time were selected according to reducing power, then the response surface method was used to determine the optimum enzymatic process. The results showed that alkaline protease had a better antioxidant activity in the two-step enzymolysis, the optimum enzymolysis conditions were as follows: pH of 8.96, temperature of 49.3℃, time of 5.7 h. Under these conditions, the reducing power of peptides reached to 13.48×10^-2, which increased 3.51% than before.
