金针菇非食用部位水提物的膜分离及体外抗肿瘤活性

The Membrane Separation of Polysaccharide from Inedible Parts of Flammulina velutipes and Characterization of Its Anti-tumor Activity in vitro

摘要 以金针菇的非食用部位为原料,探讨了该部位水提物的膜分离工艺及其体外抗肿瘤活性筛选。实验结果表明,金针菇非食用部位的水提物经过膜分离纯化后,多糖含量从9.46%提升至17.24%,得率11.30%;该部位的水提取物及其膜分离后各组分对人结肠癌(HCT-8)、人肝癌(Hep G2)、人胃癌(BGC-803)和人鼻咽癌(KB)肿瘤细胞都具有良好的抑制作用,尤其对于Hep G2和KB的作用最为明显;该部位水提取物对Hep G2和KB的IC50分别为6.9和7.6μg/m L,而膜分离后的超滤膜截留液组分抑制作用最强,对Hep G2和KB的IC50分别为1.0和0.8μg/m L。 In this study, the membrane separation process of polysaccharide was processed with inedible parts of Flammulina velutipes as raw material, and then the anti-tumor activity of polysaccharide was assayed in vitro by MTT. The results showed that, after membrane separation, the purity of polysaccharide was nearly doubled from 9.46% to 17.24%, and the yield is 11.30%. Colon cancer cells (HCT-8), human liver cancer cells (Hep G2), human gastric cancer (BGC-803) and nasopharyngeal carcinoma (KB) cancer ceils were used to test the anti-cancer ability by different polysaccharide parts separated from Flammulina velutipes, and the inhibition rate was more obvious for Hep G2 and KB, the IC50 of Hep G2 and KB were 6. 9 and 7.6 μg/mL, respectively, and the polysaccharide parts with trapped fluid of uhrafihration membrane has the strongest inhibition rate, and the IC50 of Hep G2 and KB were reduced to 1.0 and 0. 8 μg/mL, respectively.
