Effect of allicin on spleen and cell cycle of splenocytes in mice

摘要 目的利用流式细胞仪研究大蒜素对小鼠脾脏及其细胞周期的影响。方法将80只昆明小鼠随机分为高剂量900 mg/(kg·bw)、中剂量600 mg/(kg·bw)、低剂量300 mg/(kg·bw)和生理盐水对照组,各剂量组雌雄各半,每日灌胃大蒜素,对照组灌胃生理盐水。每天灌胃1次,灌胃容量为0.1 m L/10 g,连续灌胃14 d。于灌胃结束1周后将小鼠处死,分离脾脏计算脾脏器系数并制取单细胞悬液,应用流式细胞仪检测分析小鼠脾脏的细胞周期。结果高剂量组小鼠的脾脏器系数较生理盐水对照组显著升高(P<0.05);其中高剂量组和中剂量组脾G2/M期细胞、S期细胞百分比明显增加(P<0.05),脾G0/G1期细胞百分比明显减少(P<0.05),并且随着大蒜素剂量的增加,3个剂量组进行两两比较,差异有统计学意义,呈现剂量依赖性。结论大蒜素可促进脾脏产生、释放免疫活性物质,促进脾脏的生长,调节脾脏的免疫活动,使G0/G1期细胞百分比减少,G2/M期细胞、S期细胞百分比增加,缩短细胞周期,增加小鼠脾脏指数,对小鼠的免疫具有正向调节作用。 Objective To determine the effect of allicin on the spleen index and cell cycle of splenocytes in mice.Methods A total of 80 Kunming mice were randomly divided into high-,middle- and low-dose allicin groups(900,600 and300 mg/kg)and control group(normal saline). The mice of the above groups were given allicin at corresponding doses or normalsaline through intragastric gavage,once per day,0.1 m L/10 g,for 14 d. In 1 week after the end of gavage,the mice weresacrificed to collect the spleen and to make single cell suspension. Then the spleen index was calculated,and the cell cycle ofthe splenocytes was analyzed by flow cytometry. Results High- dose allicin group resulted in significantly higher spleenindex when compared with control group(P〈0.05). There were increased proportions of cells arrested in G2/M phase and Sphase(P〈0.05),and decreased cells in G0/G1phase(P〈0.05)in the high- and middle-dose groups. The effect of allicin on thecell cycle was in a dose-dependent manner. Conclusion Allicin can promote the produce and release of immunoreactivesubstances from the spleen,improve the growth of spleen,regulate spleen immune activity,decrease the cell percentage in G0/G1 phase and increase the cells in G2/M and S phase,shorten cell cycle,and increase the spleen index. Thus,allicin exerts apositive moderating effect on the immune function in the mice.
