三黄洗剂对湿疹模型动物消炎止痒的作用机制

Anti-inflammatory and anti-pruritic effects of Sanhuang Lotionon on rat model of eczema and their mechanisms

目的探讨三黄洗剂对湿疹模型动物的治疗作用。方法按体重将小鼠随机分成对照组,模型组,哈西奈德组和三黄洗剂高、中、低浓度组。将制备好的三黄洗剂对二硝基氯苯、二甲苯、组胺所致的湿疹动物模型外用处理,并利用小鼠、豚鼠双重动物模型以观察其治疗效果。结果与对照组相比,模型组小鼠在第1、2、3、4次二硝基氯苯激发后的24、48、72 h耳部均明显肿胀(P<0.05);与模型组相比,三黄洗剂低、中浓度能明显抑制小鼠耳肿胀(P<0.05);三黄洗剂高浓度组在4次激发后均极明显提高小鼠耳肿胀度(P<0.01)。三黄洗剂低、中浓度能明显抑制二甲苯所致的小鼠耳肿胀,能减轻耳片肿胀度,降低耳肿胀率,有较好的降低耳毛细血管通透性的作用(P<0.05)。三黄洗剂能抑制组胺所致豚鼠的皮肤瘙痒的作用(P<0.05)。结论三黄洗剂可以抑制小鼠耳肿胀,有抗迟发性炎症和抗急性炎症的作用;对高组胺引起豚鼠瘙痒有止痒作用。

Objeclive To determine the therapeutic efficiency of Sanhuang Lotion（SHL）on the treatment of animalmodel of eczema. Methods According to the body weight,the mice were randomly divided into control group,model group,halcinonide group and SHL high-,medium- and low-dose groups. The prepared SHL was used to treat nitrochlorobenzene,xylene,and histamine caused mouse model of eczema at the corresponding doses. Guinea pigs were employed to determine thetherapeutic effect of SHL. Results Compared with the control group,the mice of the model group displayed markedly swollenears in 24,48 and 72 h after nitrochlorobenzene challenge for 1,2,3 and 4 times（P〈0.05）. Low- and middle-dose SHLtreatment resulted in significant attenuation of the ear swelling when compared with the mice in the model group（P〈0.05）.High-dose SHL could significantly inhibit ear swelling in the mice caused by 4 times＇ stimulation of dimethylbenzene（P〈0.05）,reduce the severity of ear swelling,decrease the rate of ear swelling,lower blood capillary permeability of the ear（P〈0.05）,and SHL could remarkably inhibit the itchy skin in the guinea pigs caused by histamine（P〈0.05）. Conclusion SHLcan significantly inhibit ear swelling in the mice model of eczema,exert anti-late-onset and-acute inflammatory effects,andrelieve itching in the guinea pigs caused by histamine.
