鼻咽癌患者中Testin的表达与临床病理特征的关系 被引量:1

Relationship between Testin Expression and Clinicopathological Characteristics in Nasopharyngeal Carcinoma Patients

摘要 目的:探讨人鼻咽癌中候选抑癌基因Testin的表达,并分析其与鼻咽癌临床病理特征的关系。方法:选取治疗前经过病理确诊为鼻咽癌的患者80例活检标本作为观察组,同期选80例慢性炎症患者鼻咽黏膜组织作为对照组,采用TR—PCR和免疫组化SP分别检测两组患者的TestinmRNA表达和Testln蛋白表达,分析鼻咽癌患者TestinmRNA表达和Testin蛋白表达与临床病理特征关系。结果:鼻咽癌肿瘤标本中TestinmRNA表达和Testin蛋白表达分别为36.25%和31.25%,明显低于正常组织中的90.0%和86.25%,差异均有统计学意义(P〈0.05);鼻咽癌患者的TestinmRNA表达与性别、年龄等无关(P〉0.05);与肿瘤的分化程度、Dukes分期、淋巴结转移、远处转移等有关(P〈0.05);鼻咽癌患者的Testin蛋白表达号I生别、年龄等无关(P〉0.05),与肿瘤的分化程度、Dukes分期、淋巴结转移、远处转移等有关(P〈0.05);鼻咽癌患者TestinmRNA表达与Testin蛋白表达呈正相关关系(r=0.814,P〈0.05)。结论:鼻咽癌患者的Testin表达量明显下降,可能与鼻咽癌的分化程度、Dukes分期、淋巴结转移、远处转移等有关,提示Testin基因表达下调可能促进鼻咽癌的发生、发展。 OBJECTIVE: To investigate the expression of candidate tumor suppressor gene Testin in human nasopharynged carcinoma and to analyze the relationship between Testin expression and clinicopathological characteristics. METHODS: 80 patients with nasopharynged carcinoma were selected as the observation group and 80 patients with chronic inflammatory nasopharynged mucosa tis- sue as the control group. The mRNA SP expression and Testin protein expression were detected by TR - PCR and immunohistochemis- try Testin in two groups. And the relationship between mRNA Testin expression and Testin protein expression and cliniced pathological features were analyzed. RESULTS: mRNA Testin expression and Testin protein expression in the observation group were 90% and 36. 25% respectively, which was lower than that in the normal tissues significantly. There was statistically significant difference (P 〈 0.05). mRNA Textin expression in the patients with nasopharyngeal carcinoma was not related to gender and age ( P 〉 0.05 ), but related to tumor differentiation, Dukes stage, lymph node metastasis, distant metastasis and so on (P 〈 0.05 ). Testin protein expression in the patients with nasopharyngeal carcinoma was not related to gender and age (P 〉 0. 05), but related to tumor differentiation, Dukes stage, lymph node metastasis, distant metastasis and so on (P 〈 0.05). The expression of mRNA Testin in nasopharyngeal carcinoma patients was positively correlated with the expression of Testin protein ( r = 0. 814,P 〈0.05). CONCLUSION: The expression of Testin in the patients with nasopharyngeal carcinoma is significantly decreased, which may be related to the differentiation degree, Dukes stage, Lymth node metastasis and distant metastasis. This suggests that Testin gene expression may promote the occurrence and development of nasopharynged carcinoma.
