不同取样方法对苜蓿雄性不育系Ms-4RNA质量的影响 被引量:1

EFFECT OF DIFFERENT SAMPLING METHODS ON THE EXTRACTION OF RNA FROM ALFALFA MALE STERILE LINE MS-4

摘要 本文利用试剂盒法研究了冰盒保存不同时间对苜蓿雄性不育系MS-4花蕾和小花的RNA质量影响,拟筛选出适宜苜蓿雄性不育系MS-4 RNA研究的有效取样方法,解决远距离携带液氮取材困难的问题。结果表明:苜蓿雄性不育系MS-4花蕾的和小花RNA取样后冰盒保存6h内,琼脂糖凝胶电泳检测苜蓿雄性不育系MS-4总RNA,条带清晰,无拖尾现象,28s与18s的比例约为2:1,A_(260)/A_(280)的值在1.90-2.15,能保证提取的RNA的完整性,可作为苜蓿雄性不育系MS-4分子生物学研究的有效材料。因此,苜蓿雄性不育系MS-4花蕾和小花冰盒保存6h是基地取样短途运输的允许时间。 In this paper, the effect of different storage time in ice box on RNA quality male sterile lines MS -4's buds and flowers by kit method was studied. It will select effective sampling method for RNA research of male sterile lines MS -4 and to solve the problem of the difficulty of carrying liquid nitrogen in a long distance. The results showed that RNA which storage in ice box for 6 hours has clear bands, no tail phenomenon, 28s and 18sratio around 2:1, A260 / A280 value between 1.9 -2.15, it can guarantee the integrity of RNA. The sample which storage in ice box for 6 hours can become an effective material for molecular biology study of MS - 4. Therefore, storage in ice box for less than 6 hours is suitable time for sampling of alfalfa male sterile lines MS - 4 in short dis- tance transport.
