摘要
目的运用中医辨证论治,对疾病进行整体分析并作理论指导,探讨归脾安神颗粒治疗失眠的临床疗效,更进一步的深入了解归脾安神颗粒发挥药效的具体作用机制。方法选用清洁的wistar大鼠,并经过处理后制成失眠大鼠模型,然后将失眠大鼠模型随机分成失眠模型组、归脾安神颗粒低、中、高3个剂量组、阳性对照(枣仁安神胶囊)组,并设置空白对照组,各18只。观察戊巴比妥钠翻正实验对大鼠睡眠潜伏期及睡眠总时间的影响。采用酶联免疫法观察激素PGD2的含量,采用荧光定量PCR检测PGD2 mRNA、L-PGDS mRNA、DPR mRNA的表达。结果归脾安神颗粒可增加失眠大鼠模型的总睡眠时间,并且可以提高失眠模型大鼠下丘脑PGD2含量及PGD2 mRNA、L-PGDS mRNA、DPR mRNA的表达的作用。结论归脾安神颗粒能够改善失眠模型大鼠的睡眠,延长总的睡眠时间。药效发挥机理基本确认为通过干预失眠模型大鼠下丘脑激素而发挥改善睡眠的作用。
Objective To the theory of traditional Chinese medicine( TCM) as the foundation,study of sleeps on insomnia therapeutic effect and to further clarify the mechanism of sleeps decoction in the treatment of insomnia.Methods Wistar rats of clean grade give intraperitoneal injection of PCPA,reproduce the rat model of insomnia,will create the successful model rats were randomly divided into insomnia model group,low sleeps,and high dose group,positive drug control drug group( Zaorenanshen capsule),also set up a blank control group,18 rats in each group. The general state of rats was observed,and the difference of sleep latency and total sleep duration was observed by using sodium salt in the rats. The expression of mRNA PGD2,mRNA DPR,mRNA L-PGDS and PGD2 were detected by PCR assay. Results The general state party sleeps can improve insomnia rats and reduce the sleep latency increased total sleep time. And the expression of mRNA L-PGDS,mRNA DPR and mRNA PGD2 in the hypothalamus of the rats with insomnia can be improved. Conclusion Guipi Anshen granule can improved model of insomnia rat general state as well as reduce the sleep latency and increasing the total sleep time. At the same time,it can improve the expression of mRNA PGD2,mRNA L-PGDS,mRNA PGD2 in hypothalamus of rats with insomnia.
