α-细辛脑原位凝胶对卵清蛋白致大鼠哮喘影响的研究 被引量:3

摘要 目的:研究α-细辛脑原位凝胶穴位注射对卵清蛋白致大鼠支气管哮喘的作用机制。方法:雄性大鼠按体重随机分为正常组,模型组,羊肠线埋线组(4cm/只),空白原位凝胶组(25mg/kg),α-细辛脑原位凝胶组(25mg/kg),实验第1d和第8d分别通过腹腔、两侧腹股沟及两前足跖注射抗原液,每处0.2ml,2次致敏;第15d起用1%OVA生理盐水溶液超声雾化30min诱喘,每天1次,连续雾化一周复制大鼠哮喘模型。各组大鼠末次激发24h后,股动脉采血和取肺组织。用四川大学图象图形研究所Biomias2001图象分析系统,对肺组织HE切片进行嗜酸性粒细胞及浆细胞计数;ELISA测定大鼠血清中免疫球蛋白E(Ig E)、特异性免疫球蛋白E(s Ig E)、γ干扰素(IFN-γ)、白细胞介素4(IL-4)的表达情况。结果:α-细辛脑原位凝胶(25mg/kg)穴位埋植能明显改善模型大鼠肺的病理情况,降低血清中Ig E、s Ig E、IL-4的含量,升高IFN-γ的含量。结论:α-细辛脑原位凝胶(25mg/kg)穴位埋植对支气管哮喘具有较好的治疗作用,为治疗支气管哮喘提供新的给药方式和途径。 Objective: To study the mechanism of acupoint injection alpha asarone in situ gel in ovalbumin induced of bronchial rats asthma . Methods : 50 male SD rats were divided into five groups : control group, Model group, Catgut Embedding group, Blank in situ gel group, alpha asarone in situ gel group. We established asthmatic rat model by injected OVA and Al (OH) 3 saline suspension, at 1 days and 8 days through the abdominal cavity, and the two sides of the groin inject antigen solution per point 0.2ml, total 2 sensitizations, furthermore, at 15 days 1% OVA physiological saline solution ultrasonic nebulizer 30min induced asthma, once a day for one week. All rats were stimulated bythe last 24h,Femoral artery blood sampling and lung tissue. Counts of eosinophils and plasma ceils in HE sections of lung tissue used by Bio- mias2001 image analysis system Institute of image and graphics, Sichuan University. ELISA was used detect expression of Serum IgE, sIgE and IFN- gamma,IL-4 levels. Results: The pathological examination asthma model to replicate the success obtained; alpha asamne in situ gel group (25mg/kg)points implants can significantly improve pathological conditions rat lung model, reduce serum IgE,sIgE,IL-4 content in- creased IFN-γ, in content. Conclusion: alpha asarone in situ gel group(25mg/kg) acupoint embedding of bronchial asthma with good thera- peutic effect and provide new drug delivery methods and approaches for the treatment of bronchial asthma.
