摘要
目的:观察葡萄籽原花青素对自发性高血压大鼠(SHR)左室重塑的保护作用并探讨其机制。方法:8周龄雄性SHR24只随机分成4组,每组6只:模型组、葡萄籽原花青素50mg/kg剂量组和200mg/kg剂量组、卡托普利组(30mg/kg)。6只同龄雄性Wistar-Kyoto大鼠设为对照组。治疗8周后,测定尾动脉收缩压(SBP);观察大鼠左室结构,测量左室后壁厚度;Masson染色下观察心肌血管周围胶原纤维情况并测量心肌血管周围胶原面积(PVCA);生化法和ELISA法分别检测左室心肌组织中NO和Ang II含量;Western blot测定心肌组织中ET-1蛋白表达。结果:葡萄籽原花青素能显著降低SHR尾动脉SBP,减轻左室壁增厚,减少心肌组织胶原、Ang II含量和ET-1蛋白表达,增加NO含量。葡萄籽原花青素200mg/kg剂量组的作用尤为显著。结论:葡萄籽原花青素对SHR左室重塑有减轻作用,可能与葡萄籽原花青素降低尾动脉SBP、减少Ang II含量、ET-1蛋白表达和增加NO产生有关。
Objective: To observe the protective effects of grape seed procyanidins on the left ventricular remodeling in spontaneously hypertensive rats (SHR) and discuss its mechanism. Methods: 24 male SHR (8-week-old) were randomized into 4 groups (n = 6 each group) : SHR, grape seed procyanidins 50 and 200 mg/kg dose group, captopril used for positive control group (30 mg/kg), meanwhile, 6 Wistar Kyoto rats (8-week-old) were randomly served as control group. After 6 weeks treatment, the dynamic tail systolic pressure, the structure of left ventricular and the left ventricular posterior wall thickness were observed or were calculated. Perivascular collagen fibers and perivascular cir- cumferential collagen area (PVCA) were observed and measured by Masson staining. Biochemical techniques and ELISA were performed to determine the NO and AngII respectively while Western blot was used to detect the expression of ET-1 in myocardial tissue. Residts: grape seed procyanidins can significantly decrease tail systolic pressure and attenuate the thickness of left ventricular posterior wall, reduce perivas- cular collagen fibers, contents of AngII and expression of ET-1 but increase NO contents in myocardial tissue in SHR. These effects were more notable in grape seed procyanidin 200 mg/kg dose group. Conclusion : grape seed procyanidins possess protective effect against the left ventrictflar remodeling in SHR, this may be related to lower the tail systolic pressure, reduce contents of AnglI and expression of ET-1 but el- evate contents of NO in myocardial tissue.
