摘要
目的:研究红花黄色素对神经根型腰椎病大鼠抗炎作用及其机制。方法:建立神经根型腰椎病大鼠模型,随机分为假手术组、模型组、PDTC(吡咯烷二硫代甲酸铵盐)组、红花黄色素100 mg/kg、50 mg/kg、25 mg/kg剂量组。红花黄色素各组于造模后3 d静脉注射给药,共给药7 d。检测后肢热痛刺激缩爪阈值的变化;椎板法检测血液流变学指标;Elisa法检测大鼠血清中TNF-α、IL-1β、IL-6、COX-2水平;Western法检测脊髓背角中p-NF-k B P65蛋白的表达;RT-PCR法检测NF-k B P65 mRNA的表达。结果:与模型组相比,100 mg/kg、50 mg/kg、25 mg/kg剂量的红花黄色素均能显著提高热痛刺激缩爪阈值、改善血液流变学各指标;显著降低TNF-α、IL-1β、IL-6、COX-2水平;显著抑制p-NF-k B P65蛋白及NF-k B P65 mRNA的表达。结论:红花黄色素可能对神经根型腰椎病的治疗有一定的意义,其机制与抑制NF-k B P65通路的激活有关。
Objective: To observe the anti-inflammatory effect and mechanism of saffower yellower on lumbar spondylopathy of nerve root type in rats. Methods: The lumbar spondylopathy of nerve root type in rats was builded. Rats were randomly divided into sham, model, inhibitor (PDTC) and the 100 mg/kg, 50 mg/kg, 25 mg/kg dose of SY group. 3 days after model building, SY was administrated by intravenous in- jection, 14 days continuously. Thermal withdrawal latency was detected by therml allodynia. Hemorheology indexes were detedcted by rotary cone-plate viscometer. TNF-α, IL-1 β, IL-6 and COX-2 level in serum was detected by Elisa method, p-NF-kB P65 protein level in spinal dorsal horn was detected by western blot. NF-kB P65 mRNA level in was detected by RT-PCR. Results: Compared with model group, hemotheology indexes were improved significantly, Thermal withdrawal latency, TNF-α, IL-1β, IL-6, COX-2 level, p-NF-kB P65 protein and NF-kB P65 mRNA level was significantly decreased in the 100 mg/kg, 50 mg/kg and 25 mg/kg dose of SY(p 〈0.01 ,p 〈0.05). Conclu- sion: Saffower yellower plays an important role on lumbar spandylopathy of nerve root type and the mechanism may be related with inhibiting activation of NF-kB P65 pathway.
