大黄黄芪不同配伍比例对慢性肾衰竭大鼠24hUPQ、Scr、BUN及肾脏形态学的影响 被引量:22

Effects of different compatibilities of Rhubarb and Astragalus on 24 hour urinary protein,serum creatinine,urea nitrogen and renal tissue of rats with chronic renal failure

摘要 目的:探讨大黄黄芪不同配伍比例对慢性肾衰竭大鼠24h尿蛋白定量、血清肌酐和尿素氮以及肾脏形态学的影响。方法:SD大鼠采用5/6肾切除法建立慢性肾功能衰竭模型,随机分为正常组、假手术组、模型组、贝那普利组、大黄黄芪(1:1、1:4、1:6)组,连续灌胃给药40天,收集24h尿液量测定24h尿蛋白并测定血清肌酐和尿素氮浓度,光镜下观察肾组织并对病变进行分类评分。结果:1大鼠5/6肾切除后90d,慢性肾衰大鼠模型成功;2大黄黄芪(1:1)、大黄黄芪(1:4)和大黄黄芪(1:6)能明显减低慢性肾衰大鼠24h尿蛋白定量、血清肌酐和血清尿素氮水平,改善慢性肾衰大鼠的肾小管空泡变性和炎性坏死程。3相对于大黄黄芪(1:6)组,大黄黄芪(1:1)组降低大鼠血清Scr和BUN的作用具有统计学意义(P<0.05),肾脏病变改善效果最好。结论:大黄黄芪配伍能有效改善慢性肾衰大鼠蛋白尿的情况,增强肾小球的滤过功能,减轻肾脏病变程度,以大黄黄芪(1:1)组作用显著。 Objectives: To explore the effects of 24 hour urinary protein(24hUPQ), serum creatinine (Scr), urea nitrogen(BUN) and renal tissue of rats with chronic renal failure(CRF) treated with different combinations of Rhubarb and Astragalus. Methods: SD rats were randomly di- vided into normal control group, sham operation group, model group with CRF, Benazepril group, Rhubarb and Astragalus compatibility ( 1 : 1, 1:4, 1:6) group, then rats model of CRF were established by 5/6 nephrectomy. 40 days later, the rats were killed and the kidneys were harvested for hematoxylin - eosin dyeing. The blood samples of every group were collected respectively to detect Scr and BUN. The renal le- sions were classified and scored under light microscope. Results: ①Mter rats with renal resection 90 days, the model of the chronic renal failure is success; ②By gavaging for 40 days, Rhubarb and Astragalus membranaeeus compatibility ( 1 : 1 ), Rhubarb and Astragalus mem- branaceus compatibility ( 1:4) and Rhubarb and Astragalus membranaceus compatibility (1:6) groups decreased the level of 24h urine pro- tein, serum urea nitrogen, serum crestinine significantly of the rats of the chronic renal failure, and renal tissue lesions and inflammatory nec- rosis degree of different compatibility of Rhubarb and Astragalus groups were mitigated. ③Relative to Rhubarb and Astragalus membranaceus compatibility (1:6)group, Rhubarb and Astragalus membranaceus compatibility (1:1)group reduced the effect of rat serum urea nitrogen and serum crestinine with statistical significance ( DP 〈 0.05 ), kidney changes to improve the best effect. Conclusion: Compatibilities of Rhubarb and Astragalus increased glomerular fihration function, and significantly attenuated renal damage of rats with chronic renal failure. The effect of Rhubarb and Astragalus membranaceus compatibility ( 1 : 1 ) was most significant.
