五子衍宗丸对少弱精子症模型大鼠睾丸组织Catsper1蛋白及其mRNA表达影响 被引量:9

Effect of Wuzi Yanzong Pill on Protein and mRNA Expression of Catsper1 in Testicular Tissue of Rats with Oligoasthenozoospermia

摘要 目的通过检测大鼠睾丸组织Catsperl蛋白及其mRNA表达水平,探讨五子衍宗丸治疗少弱精子症的作用机制。方法将体质量200-220g的雄性SD大鼠随机分成正常组,模型组,生精胶囊组,五子衍宗丸低剂量、中剂量、高剂量组。采用连续灌服雷公藤多苷8周的方法复制少弱精子症模型,模型复制成功后连续给药4周。采用Westernblot法测定Catsperl蛋白含量,采用荧光定量PCR法测定CatsperlmR—NA表达水平,采用酶联免疫吸附试验检测睾丸组织中cAMP含量。结果生精胶囊和高剂量五子衍宗丸能明显提高模型大鼠睾丸组织中降低的cAMP含量以及升高睾丸组织中降低的Catsperl蛋白及其tuRNA表达水平(P〈0.05)。结论促进Catsperl蛋白表达并参与cAMP-Ca2+正反馈可能是五子衍宗丸提高精子质量的机制之一。 Objective To investigate the mechanism of action of Wuzi Yanzong Pill in the treatment of oligoasthenozoospermia through the measurement of protein and mRNA expression of Catsperl in testicular tissue in rats. Methods Male Sprague-Dawley rats with a body weight of 200-220 g were randomly divided into normal group, model group, Shengjing Capsule group, and low-, medium-, and high-dose Wuzi Yanzong Pill groups. The rat model of oligoasthenozoospermia was established by the treatment with tripterygium glycoside by gavage for 8 consecutive weeks, and after the model was successfully established, the drugs were administered for 4 consecutive weeks. Western blot was used to measure the content of Catsperl protein, fluorescence quantitative PCR was used to measure the mRNA expression of Catsperl, and enzyme-linked immunosorbent assay was used to measure the content of cAMP in testicular tissue. Results The Shengjing Capsule group and the high-dose Wuzi Yanzong Pill group showed significant increases in the reduced content of cAMP and the reduced protein and mRNA expression of Catsperl in the testicular tissue (P〈0.05). Conclusion Wuzi Yanzong Pill improves the quality of sperms through promoting the expression of Catsperl and being involved in cAMP-Ca2+ positive feedback.
