摘要
目的探讨不同年龄Sprague Dawley(SD)大鼠肝脏功能指标的差异以及自发性肝脏组织形态学的病理性变化。方法离乳3周龄SPF级SD大鼠120只,随机分为3个实验组,每组40只,雌、雄各半。分别于动物4周龄(幼龄期)、17周龄(青春期)、30周龄(成熟期)时采大鼠颈静脉血,采血后用BACKMAN COULTER全自动生化分析仪测定大鼠肝脏功能指标;处死动物,迅速取出肝脏并去除其附着组织,用电子天平称湿重,计算脏器系数,并取肝组织做组织病理学观察。结果大鼠血清总胆固醇(CHOL)含量和碱性磷酸酶(ALP)活性随着年龄增长不断下降,谷丙转氨酶(ALT)活性随着年龄增长先降后升,17周龄(青春期)时最低。4周龄(幼龄期)大鼠血清总蛋白(TP)、白蛋白(ALB)和球蛋白(GLB)含量最低。谷草转氨酶(AST)活性各周龄间无明显变化趋势。30周龄(成熟期)雌鼠2只、雄鼠7只肝脏出现轻度脂肪样变。结论 SD大鼠的部分肝脏功能指标以及自发性肝脏组织形态学改变发生率存在周龄差异。
Objetive To study the variation of hepatic function and histomorphology for the Sprague Dawley(SD) rats in the different age.Methods 120 rats of 3 weeks old were be randomly assigned into three groups by weight.Each group had 20 females and 20 males.When the animals were 4 weeks old,17 weeks and 30 weeks old,the hepatic function of blood were detcted by BACKMAN COULTER automatic biochemistry analyzer respectively.The livers of all animals should be trimmed of any adherent tissue,as appropriate,and their wet weight taken as soon as possible after dissection.Histopathology should be performed on liver tissues.Results The level of serum total cholesterol(CHOL) and the activity of alkaline phosphatase(ALP) decreased gradually as the age of the rats increased.The activity of alanine aminotransferase(ALT) at the age of 17 weeks was the lowest of the three age groups.The level of total protein(TP),albumin(ALB) and globulin(GLB) at the age of 4 weeks were the lowest of the three age groups.The activity of aspartate aminotransferase(AST) was no obvious difference during the three age groups.The livers of 2 female rats and 7 male rats were mild lipoid degeneration at the age of 30 weeks.Conclusion Some markers of hepatic function and the incidence of the change of histomorphology are different between female and male SD rats at different ages.
