中电导钙激活钾离子通道在乳腺癌中的表达及其与临床病理指标的关系 被引量:1

Expression of intermediate-conductance calcium-activated potassium channel SK4 in breast cancer and relative clinicopathological factors

摘要 目的探讨中电导钙激活钾离子通道(Intermediate-conductance calcium-activated potassium channel,SK4)在乳腺癌组织和细胞系中的表达及其临床意义。方法 42例乳腺癌组织标本,20例距肿瘤切缘2 cm的癌旁组织标本。免疫组织化学法(Immunohistochemistry,IHC)和Western Blot法(WB)检测SK4在乳腺癌组织和乳腺癌细胞系中的表达,并分析其与临床病理指标的关系。结果 SK4在癌组织中的表达高于癌旁组织。SK4在乳腺癌组织和癌旁组织中的表达阳性率分别为88.1%和35.0%(P<0.05);低、中、高分化肿瘤中SK4表达阳性率分别为100.0%、87.5%和75.0%(P<0.05);伴或不伴淋巴结转移的患者SK4的表达阳性率分别为69.2%和99.6%(P<0.05);在乳腺癌雌激素受体阳性和阴性中SK4的阳性率别为86.7%和91.7%(P>0.05)。SK4在乳腺癌细胞系MDA-MB-231和MCF-7中明显表达。结论 SK4在乳腺癌组织中表达增加;SK4的表达与肿瘤分化程度、淋巴结转移相关。 Objective To study the expression and significance of intermediate-conductance cal-cium-activated potassium channel SK4 in breast cancer tissues and cell lines. Methods Immunohisto-chemistry(IHC)staining and Western Blot(WB)was performed to detect the expression of SK4 in 42 ca-ses of breast cancer tissues and breast cancer cell lines. The relationship between SK4 and relative clinico-pathological factors was also studied. Results The positive rates of SK4 in cancer tissues and adjacent tissues were 88. 1% and 35. 0% . The expression rates of SK4 in poorly-differentiated,median-differentia-ted and well-differentiated tumors were 100% ,87. 5% and 75. 0% ,respectively. The expression rates of SK4 were 69. 2% and 99. 6% in patients with lymphatic metastasis and patients without it. The expression rates of SK4 in ER-positive group and ER-negative group were 86. 7% and 91. 7% ,respectively. Conclu-sion SK4 expression is increased in breast cancer. The SK4 expression rate is related to clinicopathologi-cal factors including tumor differentiation and lymph node involvement,suggesting SK4 maybe a potential candidate for targeting therapy.
