摘要
以禾本科牧草高羊茅为研究对象,采用液相色谱电离串联质谱(LC-ESI-MS)分析了干旱胁迫条件下高羊茅叶片中的代谢组学变化。其中,共稳定检测到282种代谢产物发生变化,148种下调表达,134种上调表达。利用MZmine软件处理原始数据,用主成分分析(PCA)与正交偏最小二乘法判别分析(OPLS-DA)方法,鉴定出4种代谢产物在干旱处理后下调,6种代谢产物上调。其中,芳香族化合物酰基基诺内酯、芳香族游离氨基酸、苏合香脂、甲烷呋喃都表现为上调;油脂代谢产物氢过氧化硬脂丁二酸酐、3-氧代十二烷酸酯、3-羟基十八烷基碳稀酸表现为下调,过氧化三甲基胺甲脂表现为上调;其他代谢产物如羟甲基茉莉酸葡萄糖苷(糖苷类)上调,胡椒新碱(生物碱)下调。芳香族化合物和脂代谢产物可能在高羊茅抗旱过程中起关键调控作用。
The metabolites were analyzed with LC-ESI-MS(liquid chromatography tandem mass spectrometry)in tall fescue leaves under normal and drought conditions.A total of 282 metabolites were stably detected in the two groups using OPLS-DA(orthogonal partial least squares discriminant analysis)method;148of which were down-regulated while 134 were up-regulated.After analyzed with MZ-mine software in raw data,10 major metabolites(4down,6up)were identified through searching public database.They were aromatic compounds:3-p-coumaroyl1,5-quinolactone,aromatic free amino acids,styraxin,Methanofuran;Grease metabolites:methyl 15-hydroperoxy-9Z,12 Z,16E-octadecatrienoate,9,12,13-TriHOME(10),3-oxo-dodecanoic acid,13(S)-HpOTrE acid ester;and other metabolites:Piperanine(alkaloids)and Methyl 7-epi-12-hydroxyjasmonate glucoside(indican).In a word,aromatic compounds and the lipid metabolites may play vital roles in tall fescue drought-resistant regulation.
