心抑瘤素M及其受体在压力负荷致心肌肥厚小鼠心肌组织中的表达及意义

摘要 目的探讨心抑瘤素M(OSM)/OSM受体(OSMR)在压力负荷致心肌肥厚小鼠心肌组织中的表达及其意义。方法选择野生型C57BL/6J小鼠60只,随机分成为假手术组(Sham组)、模型组(AB组)各30只,AB组采用胸主动脉缩窄术建立心肌肥厚模型。两组于术后4周行超声检测心功能,处死后观察心脏大体观并行心肌组织病理学检测。应用RT-PCR检测小鼠心肌组织OSM、IL-31、OSMR mRNA表达,应用Western blotting检小鼠心肌组织测OSM、IL-31、OSMR亚单位(OSMRβ)蛋白表达。结果术后4周,大体观察显示AB组小鼠心脏体积增大,心腔扩大,病理染色显示心肌细胞横截面积增加。与Sham组相比,AB组舒张期室间隔厚度、左室舒张末内径和左室收缩末内径增大,射血分数及短轴缩短率减小(P均<0.05);AB组心肌OSM mRNA、OSM蛋白、OSMR mRNA、OSMRβ蛋白表达均较Sham组明显上调(P<0.05或<0.01)。结论压力负荷致心肌肥厚模型小鼠心肌组织中OSM及其OSMRβ均发生异常表达,OSM/OSMRβ系统参与了心肌重构导致心肌肥厚的过程。
