摘要
为了解天鹅湖湿地微生物群落结构组成及动态变化,本研究对不同季节环境因子进行了检测,并采用PCR-DGGE技术分析了天鹅湖湿地青龙湖位点不同季节微生物群落结构多样性。环境因子检测结果表明:天鹅湖湿地p H值介于6.76~6.89之间,总氮含量范围在3.4~5.8 mg/kg,9月总氮含量最高,达到5.8 mg/kg,12月含量最低,为3.4 mg/kg。总磷含量差异不大,范围介于0.14~0.19 mg/kg,其中9月份相对较高,达到0.19 mg/kg。PCR-DGGE结果显示:3个季节样品中微生物群落结构较为健全,物种丰富度高,群落结构相似性在76.9%~89.3%之间,季节间微生物群落结构存在差异。DGGE条带测序显示湿地优势类群与α-变形菌门(Alpha Proteobacterium)相似性最高,达到98%。
In this study, in order to understand the structure composition and dynamics of microbial community in Swan lake wetland, environmental factors and microbial community composition and diversities of Swan lake wetland were analyzed in May, September and December using PCR-DGGE method, respectively. The detection results of environmental factors showed that the pH value was between 6.76 and 6.89, the contents of total nitrogen were 3.4-5.8 mg/kg, the highest and the lowest content in September and December were 5.8 mg/kg and 3.4 mg/kg, respectively. The content of total phosphorus was 0.14-0.19 mg/kg, which almost keep constant. PCR-DGGE result showed that the microbial community composition and diversity were rich, similarity of community composition between 76.9% and 89.3%, and the difference of community composition was obvious among three seasons. The sequence results of DGGE band indicated that the dominant group of wetland has the highest affinity with Alpha Proteobacterium, and their sequence comparability was 98%.
