利用SSR与RAPD分子标记评估甘蔗品种的遗传多样性 被引量:47

Genetic Diversity Evaluation of Sugarcane Varieties Using SSR and RAPD Markers

摘要 利用SSR与RAPD两种分子标记对美国、中国台湾以及中国大陆不同甘蔗育种单位选育的甘蔗品种或亲本材料的遗传多样性进行评估。其中19对SSR引物共扩增出87条带,多态性带为84条,多态性比例为96.55%,扩增出的条带数范围为2~8条,平均每对引物扩增出4.58条带,引物的PIC值范围为0.34~0.93,平均0.64。21条RAPD引物共扩增出184条带,扩增条带数范围为3~16,平均每条引物扩增8.76条带,其中多态性带为184,多态性比例为100%,引物PIC范围为0.53~0.97,平均0.86。结果表明,两种分子标记都能较好的评估甘蔗品种的遗传多样性。 SSR and RAPD molecular markers were used in this study to evaluate the genetic diversity of sugar- cane varieties and clones bred by USA, Taiwan China and different sugarcane breeding unit in China's Mainland. A total of 87 bands were amplified using 19 pairs of SSR primers. Eighty-four of them were polymorphic bands and the polymorphic rate was 96.55%. The number of amplified bands ranged from 2 to 8, with 4.58 average amplified bands for each primer pair. The polymorphism information content (PIC) value for each SSR primer pair ranged from 0.34 to 0.93 with a mean value of 0.64. A total of 184 polymorphic bands were detected by 21 RAPD primers, and the number of bands amplified by each primer ranged from 3 to 16. There were average 8.76 bands amplified for each primer. All the 184 bands were polymorphic and the polymorphic rate was 100%. The PIC value for each RAPD primer ranged from 0.53 to 0.97 with a mean value of 0.86. The results showed that the genetic diversity of sugarcane varieties could be evaluated by both two molecular markers.
