黄芪汤对糖尿病肾病小鼠AMPK-Camkkβ(LKB1)信号通路的影响 被引量:11

Influence of Huangqi Decoction on the AMPK-Camkkβ(LKB1) Signaling Pathway in Mice with Diabetic Nephropathy

摘要 目的:探讨黄芪汤改善糖尿病肾病的作用及可能的机制。方法:40只6周龄db/db小鼠随机分为模型组及黄芪汤高、中、低剂量组,每组10只,10只6周龄db/m小鼠作为正常对照组。模型组、正常组均予0.5%羧甲基纤维素钠灌胃;各药物组分别予黄芪汤1.08 g/kg、0.36 g/kg、0.12 g/kg灌胃。1次/d,灌胃14周后进行生化指标、肾组织病理学检测,Western blot检测各组肾皮质中AMPK-Camkkβ(LKB1)信号通路相关蛋白的表达量。结果:与模型组比较,黄芪汤能降低db/db小鼠的体质量、肾质量、血清三酰甘油(TG)、胆固醇(TC)、低密度脂蛋白(LDL-C)浓度,降低血清谷氨酸转氨酶(ALT)、谷草转氨酶(AST)浓度,升高肾质量/体质量指数(P<0.05),改善肾脏病理情况;上调肾皮质中p-AMPK、p-Camkkβ、p-LKB1、p-ACC蛋白表达量(P<0.05)。结论:黄芪汤通过有效活化AMPK-Camkkβ(LKB1)信号通路改善糖尿病肾病。 Objective: To investigate the effect and possible mechanism of Huangqi Decoction (HQD) on diabetic nephropathy. Methods: Forty db/db mice of 6 weeks old were randomly divided into the model group treated with 0.5% sodium carboxymethyl and HQD groups treated with Huangqi Decoction at dose of 1.08,0.36,0.12 g/kg as high, middle and low respectively, oral administration for once a day. 10 mice were in each group, ten db/m mice of 6 weeks old were randomized into the normal control group. Biochemical determination and renal histopathology were performed after 14 weeks. The expression of related protein in AMPK-Camkkβ (LKB1) pathway in renal cortex were measured by Western blot. Results: Compared with the model group, the body weight, kidney weight, serum triglycerides ( TG), cholesterol ( TC ), low density lipoprotein( LDL-C), serum ALT and AST were decreased and the ratio of kidney and body weight were increased in HQD- treated groups ( P 〈 0.05 ). The renal pathology was improved in the HQD-trcated groups, and the expression of p-AMPK, p- Camkkβ, p-LKB1 and p-ACC in the renal cortex were up-regulated (P 〈 0.05 ). Conclusion: Huangqi Decoction can improve diabetic nephropathy through the effective activation of the AMPK-Camkkβ( LKB1 ) signaling pathway.
