摘要 建立反相高效液相色谱法,同时测定食品中VA和VE(α-生育酚)的方法。取一定量乳粉加入水、VC的乙醇溶液、氢氧化钾,于53℃条件下皂化30 min,立刻冷却,乙腈提取,提取液调至中性,进样20μL分析。选择色谱条件为C18柱色谱柱,250 mm×4.6 mm,粒径5μm,流动相为甲醇,流速1.0 m L/min,柱温35℃,检测波长325 nm。结果表明,VA在0.1~1.0μg/m L质量浓度范围内线性良好,相关系数R^2=0.999 8;VE在1.0~5.0μg/m L质量浓度范围内相关系数R^2=0.999 0;VA和VE的平均回收率在95%以上。该法前处理简单快捷、分离度高、灵敏度高且准确可靠,大大提高了实验室的工作效率。 Establish A reversed phase high performance liquid chromatography and determination of vitamin A and vitamin E in foods(alpha tocopherol) method. Powder add certain amount of water and ethanol solution of vitamin C and potassium hydroxide, 53 ℃ saponification for 30 min, cooling immediately, acetonitrile extraction, extract to neutral and sample analysis of 20 μL. Chromatographic conditions:chromatographic column C18 column,250 mm×4.6 mm,size 5 μm,mobile phase: methanol flow, rate 1.0 mL/min, column temperature 35 ℃, detection wavelength of 325 nm. The vitamin A has good linear concentration range at 0.1-1.0 μg/mL, correlation coefficient R^2=0.999 8, vitamin E has concentration range correlation as 1.0-5.0 g/mL coefficient R2=0.999 0. Vitamin A and vitamin E,the average recovery is above 95%. Before the deal with simple and quick, high degree of separation, high sensitivity and accurate and reliable, greatly improving the efficiency of the laboratory.
