RD105缺失基因检测法在结核分枝杆菌分型中的应用 被引量:2

摘要 目的探讨RD105缺失基因检测法在结核分枝杆菌分型中的应用。方法 133株结核分枝杆菌分别采用Spoligotyping分型法和RD105基因缺失法鉴别北京基因型。结果 133株结核分枝中114株为北京基因型,占85.7%,19株为非北京基因型,占14.3%,经RD105缺失基因检测133株结核分枝杆菌临床分离株中,115株为北京基因型,占86.4%,18株为非北京基因型,占13.6%,Spoligotyping基因分型法与RD105缺失基因检测法比较差异无统计学意义(χ2=0.333,P=0.563),RD105缺失基因检测法的灵敏度为99.1%,特异度为89.5%,阳性预测值为98.3%,阴性预测值为94.4%。ROC曲线下面积为0.943。结论 RD105基因缺失法检测可以替代Spoligotyping基因分型法。 Objective To identify the Mycobacterium tuberculosis detected by RD105 deletion test. Methods 133 strains of M. tuberculosis were identified with Spoligotyping testand RD105 deletion test respectively to classify the Beijing genotype. Results Of the 133 clinical isolates of M. Tuberculosis,114 strains( 85. 7%) were belonged to the Beijing genotype,and 19strains( 14. 3%) were belonged to the non-Beijing genotype identified by the Spoligotyping test. While 115 strains( 86. 4%) were belonged to the Beijing genotype and 18strains( 13. 6%) were belonged to the non-Beijing genotype identified by the RD105 deletion test. There were no significant differences between the two kinds of test( χ2= 0. 333,P = 0. 563). The sensitivity,specificity,positive predictive value,negative predictive value and ROC curve area of the RD105 deletion test were 99. 1%,89. 5%,98. 3%,94. 4%,0. 943,respectively.Conclusions RD105 deletion testcan replace the Spoligotyping test.
