摘要 目的研究IL-23R的表达及基因多态性与脓毒症引起的急性肺损伤临床预后相关性。方法选取脓毒症引起的急性肺损伤病人与健康者对照,分别抽取外周血并提取总RNA和DNA,利用PCR技术测定IL-23R的mRNA值;利用PCR结合测序技术测定IL-23R不同位点基因多态性。结果脓毒症引起的急性肺损伤病人IL-23R表达显著上调,并且对30天死亡率有较强的预后评估作用;IL-23R基因的rs10889677和rs1004819位点与30天死亡率显著相关。结论 IL-23R的表达及基因多态性与脓毒症引起的急性肺损伤预后密切相关。 Objective To study the association of interleukin- 23 receptor( IL- 23R) expression and genetic polymorphisms with sepsis-induced acute lung injury( ALI). Methods 45 cases of patients with severe sepsis and113 cases of general sepsis caused ALI and 21 cases of healthy volunteers were enrolled in this study. Plasma IL-23 R mRNA were determined by qRT-PCR. Genotyping analysis of IL-23 R was performed using PCR-followed sequencing. Results The relative plasma IL- 23 R concentrations were significantly increased in severe sepsis and sepsis-induced ALI,compared with control subjects,and the expressions were highly related with the 30-day mortality rate. rs10889677 and rs1004819 genetic Polymorphisms were associated with the 30-day mortality rate. Conclusion There is close association between IL-23 R expression and genetic polymorphisms and the prognosis of sepsis-induced acute lung injury.
