摘要 目的对大连口岸淡色库蚊抗药性进行监测分析,探索该口岸淡色库蚊杀虫剂的合理使用,减少用药的盲目性,降低环境污染,提高防治效率。方法本研究以大连口岸淡色库蚊为主要研究对象,通过野外现场采集、实验室标准化养殖、抗药性生物学测试和拟除虫菊酯杀虫剂击倒抗性分子检测等方法,比较对几种杀虫剂的敏感度,从而找到敏感度相对高的杀虫剂进行蚊类灭杀。结果对淡色库蚊及幼虫进行抗药性毒力测试结果显示,大连口岸3个地理株对溴氰菊酯敏感度相对较高,对敌敌畏的抗性最高。检测不同种群拟除虫菊酯类杀虫剂击倒抗性(kdr)基因频率,结果显示大连口岸3个地理株的淡色库蚊对拟除虫菊酯类药物已经产生抗药性。结论通过研究的各项检测结果,提出以下杀虫剂合理使用措施:大连口岸杀灭淡色库蚊应该采用敏感性较高的几种拟除虫菊酯类杀虫剂轮用,也可对其加入增效剂来使用。 Objective This article monitors and researched Culex pipiens pallens resistance at Dalian ports, in order to reduce the blindness of medication and the environmental pollution, and improve the control efficiency.Methods Culex pipiens pallens of Dalian ports is the main research object in this research. Methods are used such as field collection, laboratory standard breeding, biology resistance test, molecular detection of pyrethrum insecticide knockdown resistance. The sensitivity of Culex pipiens pallens to several insecticides is compared.The insecticide of relatively high sensitivity is discovered to kill Culex pipiens pallens. Results The resistance toxicity test for Culex pipiens pallens and larvae is conducted by immersion method. The result shows that three geographical strains of Dalian have relatively high sensitivity to deltamethrin; also they are of the highest resistance to DDVP. The method of allele specific PCR should be established to detect knockdown resistance(Kdr)gene frequency of different populations of pyrethrum insecticide. That indicates that Culex pipiens pallens of Dalian ports have generated resistance to pyrethrum insecticide. Conclusion According to each test results of the research, the rational use measures of insecticides are as follows: the high sensitivity of pyrethrum insecticide should be taken in turn to kill Culex pipiens pallens of Dalian ports, and the synergistic agent can be added to use.
