芪元颗粒中总皂苷的提取和纯化工艺研究 被引量:3

Study on extraction and purification process of total saponins from Qiyuan Granules

摘要 目的:优选芪元颗粒中总皂苷的提取和纯化工艺。方法:以出膏率和总皂苷得率作为指标,采用L9(34)正交试验确定最佳的提取工艺条件;并采用大孔树脂吸附法,以大孔树脂类型、上样量、洗脱剂种类和体积、洗脱流速为影响因素,确定最佳纯化工艺条件。结果:最佳的提取工艺为:用6倍量90%的乙醇提取2次,每次1h;最佳的纯化工艺为:采用大孔树脂D-101,上样量为20m L(浓度为21.53mg/m L),用70%的乙醇65m L进行洗脱,洗脱流速为2BV/h。结论:所得提取和纯化工艺简单可行,效果良好。 Objective: To optimize the extraction and purification process of total saponins from Qiyuan Granules. Methods: The orthogonal test was adopted to determine the optimal extraction technology, using the paste rate and the yield of total saponins as indexs; the method of macroporous resin was used to determine the optimal purification technology with the type of macroporous resin, the amount of sample, the type and volume of elution solvent and the flow rate of elution as the influence factors. Results: The most optimal extraction process was as follows: the amount of 90% ethanol with 6 times was extracted 2times for 1h each time; the most optimal purification process was as follows: the type of macroporous resin was D-101, the amount of the sample was 20 m L(the concentration was 21.53mg/m L), the flow rate of elution was 2BV/h, and eluted with 65 m L of 70% ethanol. Conclusion: The extraction and purification process is simple and feasible, and it has a good effect.
