miR-126对宫颈癌细胞增殖与侵袭的影响 被引量:2

摘要 [目的]探讨miR-126对宫颈癌细胞增殖与侵袭的影响。[方法]将宫颈癌HeLa细胞分为miR-126 mimics组(转染miR-126 mimics)、scramble组(转染scramble)、Crk—siRNA组(转染Crk-siRNA)、si—control组(转染si-control),其中si—control组为Crk—siRNA组的阴性对照,scramble组为miR-126mimics组的阴性对照。采用Western blot法检测外源过表达miR-126对宫颈癌HeLa细胞Crk蛋白表达水平的影响;采用MTT和Transwell侵袭实验检测外源高表达miR-126对宫颈癌HeLa细胞增殖与侵袭能力的影响。[结果]Western blot结果显示,miR-126 mimics组和CrksiRNA组Crk蛋白表达水平较阴性对照组(scramble组和si—control组)明显降低;MTT结果显示,转染miR-126 mimics和Crk—siRNA组HeLa细胞的增殖速度从48h起明显减慢,与各自的阴性对照组比较差异均有统计学意义(P〈0.05);Transwell结果显示,转染miR-126mimics和CrksiRNA组36h后HeLa细胞穿过基质胶的细胞数量分别为(66±10)和(63±9),与对照组(157±12)比较,差异具有统计学意义(P〈0.01)。[结论]miR-126可能通过靶向调控Crk,从而抑制宫颈癌细胞的增殖与侵袭。
