摘要
目的:观察益气健脾方对CAG大鼠血清中Reg I和PCNA表达的影响及病理组织学的变化,探讨益气健脾方对CAG大鼠成模的抑制作用及其可能机制。方法:将60只SD大鼠随机分为正常组、模型组、益气健脾方低、中、高剂量组和阳性对照组,每组10只,采用MNNG联合饥饱失常的综合造模方法制备慢性萎缩性胃炎大鼠模型,10周后观察益气健脾方对CAG大鼠模型成模的影响,并测定血清中Reg I及PCNA表达含量等的变化情况。结果:1益气健脾方不同剂量组和阳性对照组对CAG大鼠在一般情况和病理状态等方面均有改善作用。2益气健脾方各组与阳性对照组对血清中Reg I及PCNA含量具有下调作用,与模型组相比具有显著差异(P<0.05),且益气健脾方高剂量组优于阳性对照组(P<0.05),呈现出一定的剂量依赖趋势。结论:益气健脾方对CAG大鼠成模具有抑制作用,能够改善CAG大鼠胃黏膜病理萎缩状态,其机理除与传统中医功效理论和药物组成的药理作用相关,与调控血清中Reg I及PCNA表达亦相关,这可能是其发挥效应的作用机制之一。
Objective ： To observe the effect of Yiqi Jianpi Formula on RegI, PCNA and pathology and discuss the inhibition and the possible mechanism for its efficacy on CAG rats. Methods ： Sixty SD rats were randomly divided into normal group, modem medicine group, Yiqi Jianpi Formula low, middle and high dose groups and the positive control group. CAG rats model was prepared with MNNG and other factors. After 10 weeks, the inhibiting effect of Yiqi Jianpi Formula on CAG rats＇ RegI and PCNA in serum were observed. Results ： （1）There were some changes in general condition and pathology treated by the different doses of Yiqi Jianpi Formula and modem medicine. （2）The different doses of Yiqi Jianpi Formula and the modem medicine could lower the contents of RegI and PCNA（ P 〈 0.05 ） while Yiqi Jianpi Formula was better than the modem medicine group（ P 〈 0.05 ） and related to the dose. Conclusions： Yiqi Jianpi Formula can inhibit the CAG rats modeling and improve the gastric pathology. The mechanism except related to the traditional Chinese medicine theory and composition of the drug - related external pharmacology, is also related to regulating RegI and PCNA expres- sions in serum. It may be one of mechanisms to treat CAG.
