摘要
目的:探讨多种生物制剂参与三维PCL支架诱导异位骨髓模型形成以及不同制剂对骨髓形成的影响。方法:将小鼠皮下埋置含有不同生物制剂的PCL支架,八周后对活体小鼠进行Micro-CT扫描之后行安乐死。小鼠取小骨进行组织学染色、造血集落形成以及流式细胞检测Sca-1+造血干细胞。结果:(1)生物修饰PCL支架诱导皮下小骨形成。(2)小骨中具有典型骨髓结构及三系造血细胞。(3)异位骨髓中造血前体细胞集落形成。(4)小骨中存在Sca-1+造血干细胞。结论:在PCL支架上添加rh BMP-2、HA、matrigel基质胶和淫羊藿苷可在小鼠皮下诱导出类真实的骨和骨髓环境。
Objective : To discuss if three - dimensional PCL scaffolds with several biological agents may format ectopic bone marrow and the effect of different preparations on the formation of ectopic bone marrow. Methods : Mice were implanted PCL scaffolds subcutaneously with different biological agents. Eight weeks later, mice were examined by Micro - CT examination or euthanized. And then the scaffolds were harvested and processed for histological staining, hematopoietic colony formation and flow cytometry. Results : ( 1 ) The biomodification of PCL scaffold induced subcutaneously ectopic ossicles formation. (2)There were typical bone marrow architecture and three hematopoietic cells in ectopic ossicles. (3) The hematopoietic progenitor cell colony formation was within ectopic bone marrow. (4) The hematopoietic stem cells existed in the bone marrow. Conclusion: PCL scaffolds biomodified with rhBMP -2, HA, Matrigel and icariin may mimic the real environments of bone and bone marrow.
