一贯煎对卵巢早衰大鼠模型脂质过氧化状态的影响 被引量:10

Anti-Peroxidation Effects of Yiguanjian on rat model of Premature Ovarian Failure

摘要 目的:观察卵巢早衰大鼠子血清中丙二醛(MDA)、过氧化氢酶(CAT)、总超氧化物歧化酶(TSOD)的水平,探讨一贯煎制剂对卵巢早衰大鼠过氧化状态的拮抗作用。方法:实验设正常对照组、模型对照组、乙烯雌酚干预组、一贯煎干预组。灌胃给予雷公藤多苷50 mg/kg·d复制卵巢早衰大鼠模型,2周后灌胃给予各干预药,连续给药6周。第7周时检测各组大鼠的卵巢病理及卵巢指数,血清中MDA、Caspase-3、T-SOD、CAT水平。结果:比较四组卵巢病理HE染色、卵巢指数,说明造模成功,且一贯煎对卵巢早衰具有一定改善效果(P<0.05)。与正常对照组比较,模型对照组的T-SOD、CAT水平明显降低(P<0.05),MDA的水平显著升高(P<0.05);与模型对照组比较,各干预药组上述指标均有不同程度的改变(P<0.05)。结论:一贯煎制剂对卵巢早衰大鼠过氧化状态有拮抗作用,其作用机制与提高T-SOD、CAT水平;并降低MDA、Caspase-3的水平有关。 Objective : To observe the levels of serum malondialdehyde ( MDA), catalase (CAT) and total - superoxide dismutase (T - SOD) of the premature ovarian failure rats before and after taking Yiguanjian and discuss the anti - peroxidation effect. Methods : The experiment rats were divided into normal control group, model control group, diethylstilbestrol group and Yiguanjian group. Intragastric administration of tripterygium glycosides was used to induce rats models of pre- mature ovarian failure. 2 weeks later, each drug intervention was given by intragastric administration, continuously for 6 weeks. And then detect the levels of T - SOD, CAT and MDA in serum. Results: Comparing HE staining and ovarian index,it indicated the success of modeling. Yiguanjian had relieve effect on premature ovarian failure. After 6 weeks, the ovarian pathology and index were different ( P 〈 0.05 ). The levels of MDA and Caspase - 3 in the intervention groups were significantly decreased ( P 〈 0.05 ) and the levels of CAT and T - SOD significantly increased ( P 〈0. 05 ). Conclusion: Yiguanjian has anti -peroxidation effect on POF rats. The mechanism is related with increasing T- SOD and CAT levels and reducing MDA and Caspase- 3.
