摘要
采用光学生物显微镜观测结合实时荧光PCR检测方法对马蹄淀粉中掺杂行为进行定性鉴别,以期为鉴别淀粉掺杂成分提供更客观、更准确的检测手段。结果显示:32个随意掺杂的马蹄淀粉样品中8个是纯马蹄淀粉,5个是纯木薯淀粉,1个是纯玉米淀粉,3个是纯马铃薯淀粉,3个是马蹄淀粉和马铃薯淀粉的混合物,1个是马蹄淀粉和木薯淀粉的混合物,1个是马蹄淀粉和玉米淀粉的混合物,5个是木薯和玉米淀粉的混合物,3个是木薯淀粉和马铃薯淀粉的混合物,2个是木薯淀粉、玉米淀粉、马铃薯淀粉的混合物。32个样品显微镜镜检结果和PCR检测结果基本吻合。两种方法可为国内淀粉市场质量监督提供有效的检测方法。
The granular adulterated properties of water chestnut starch were investigated experimentally by using the modem analytical techniques such as light microscopy and RT-PCR assay. The results showed that eight commercialized samples were pure water chestnut starchs,five in 32 samples were pure tapioca starchs,one was pure corn starch,three were pure potato starchs,three were mixture of water chestnut starch and potato starch, one was mixture of water chestnut starch and tapioca starch, one was mixture of water chestnut starch and corn starch ,five were mixture of tapioca starch and corn starch ,three were mixture of tapioca starch and potato starch, two were mixture of tapioca starch,corn starch and potato starch.The results using microscopy was consistent with that of RT-PCR assay.Two kinds of detection methods could be used as product quality supervision of domestic starch market.
