摘要
目的采用超高效液相色谱法(UPLC)快速测定甲钴胺片的含量。方法采用Agilent Zorbax Plus C_(18)色谱柱(50 mm×3.0 mm,1.8μm),以乙腈-水为流动相,梯度洗脱,流速0.8 m L·min^(-1),检测波长267 nm,柱温30℃,采集频率80 Hz,进样体积3μL。结果甲钴胺片13.95~25.91μg·m L^(-1)与峰面积呈良好的线性关系(r=0.9999),平均回收率为100.0%,RSD=0.8%(n=9)。结论所用方法简便、准确、专属性强,可用于甲钴胺片的含量测定。
OBJECTIVE To establish a UPLC method for the content determination of Mecobalamin tablets. METHODS The sepa- ration was performed on Agilent Zorbax Plus C18 (50 mm × 3.0 mm, 1.8 μm) column. The mobile phase was acetonitrile -water with gradient elution at a flow rate of 0.8 mL·min^-1. The detection wavelength was 267 nm with the column temperature of 30℃, and the collection frequency was 80 Hz, and the injection volume was 3μL. RESULTS The linear range was 13.95 - 25.91 μg·mL^-1 ( r = 0. 9999), and the recovery rate of the method was 100.0% with the RSD of 0.8%. CONCLUSION The method is simple, accurate and selective for the determination of Mecobalamin tablets.
