白藜芦醇对肺癌A549细胞增殖及NF-κB表达的影响 被引量:8

摘要 目的观察白黎芦醇对人肺癌细胞A549增殖及核转录因子κB(nuclear factor-kappa B,NF-κB)表达的影响。方法选用噻唑蓝(methylthiazolyldiphenyl-tetrazoliumbromide,MTT)法检测白藜芦醇对A549细胞增殖的影响;采用逆转录PCR(reverse transcription-PCR,RT-PCR)检测白藜芦醇对A549细胞NF-κB信使核糖核酸(messenger RNA,mRNA)表达的作用;采用免疫印迹法检测白藜芦醇对肺癌A549细胞NF-κB蛋白水平的影响。结果随白藜芦醇浓度的增加,B、C、D组细胞生长抑制率均高于A组,C、D组高于B组,D组高于C组(P<0.05)。B、C、D组24、48h细胞生长抑制率高于12h,48h细胞生长抑制率高于24h(P<0.05)。经100μmol/L白藜芦醇作用48h后,A549细胞可以明显下调NF-κB mRNA和蛋白水平的表达。结论白藜芦醇可以明显抑制肺癌A549细胞的增殖及NF-κB的表达。 Objective To study the effect of resveratrol on proliferation of human lung cancer cell line A549 and the expression of nuclear factor kappa B(NF-κB).Methods The effect of resveratrol on the proliferation of A549 cells was detected by methylthiazolyldiphenyltetrazoliumbromide(MTT)method.The effect of resveratrol on the expression of mRNA NF-κB in A549 cells was detected by reverse transcription-PCR(RT-PCR).Effect of resveratrol on NF-κB protein levels in lung cancer A549 cells was detected by western blotting.Results With the increase of the concentration of resveratrol,the cell growth inhibition rates in group B,C and D were higher than in that in group A.The cell growth inhibition rates in group C and D were higher than that in group B.The cell growth inhibition rates in group D was higher than that in group C(P〈0.05).Cell growth inhibition rate at 24 hand 48in group B,C and D are higher than that at 12 h.Cell growth inhibition rate at 48 hwas higher than that at 24h(P〈0.05).A549 cells with 100mol/L resveratrol for 48 hcan significantly down regulate the expression of NF-kappa mRNA.Conclusion Resveratrol can significantly inhibit the proliferation of lung cancer A549 cells and the expression of NF-κB.
