Simultaneous Determination of Three Nucleosides in Hericum erinaceus Extract by HPLC

目的:建立高效液相色谱法同时测定猴头菇提取物中尿苷、鸟苷和腺苷的含量。方法:采用Welch Ultimate AQ C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.05 mol·L-1磷酸二氢钾溶液,梯度洗脱;检测波长为260 nm。结果:尿苷、鸟苷、腺苷的线性范围分别为0.034 43~0.688 6μg(r=1)、0.022 26~0.445 1μg(r=1)、0.026 93~0.538 6μg(r=0.999 9),尿苷、鸟苷、腺苷的平均回收率(n=6)分别为97.6%(RSD=2.0%)、96.3%(RSD=1.0%)、96.3%(RSD=0.8%)。结论:该方法简便、准确、可行,可为猴头菇提取物的生产和使用提供质量评价依据。

Objective： To establish an HPLC method to simultaneously determine the contents of uridine, guanosine and adenosine in Hericum erinaceus extract. Methods： Welch Ultimate AQ-C18 （250 mmx 4.6 ram, 5p^m） was used for determination. Acetonitrile -0. 05 mol· L- 1 potassium dihydrogen phosphate solution was used as the mobile phase in a gradient elution mode, and the detection wavelength was 260 nm. Results： The linearity ranges obtained from calibration curves of uridine, guanosine and adenosine were 0. 034 43 -0.688 6 μg （r=1）, 0.022 26 - 0.445 1 μg （r=1）, 0.026 93 - 0. 538 6 μg （ r = 0. 999 9 ）, respectively. The average recoveries （ n = 6 ） were 97. 6% （ RSD = 2. 0% ）, 96. 3% （ RSD = 1.0% ）, 96. 3% （RSD = 0. 8% ）, respectively. Conclusion： The method is simple, accurate and validated, which can be used for the quality evaluation of H. erinaceus extract.
