摘要
目的:探究饮用不同浓度的白酒对单纯性肥胖大鼠肝脏功能的影响。方法:选取30只SD大鼠随机分为5组:对照组、单纯性肥胖模型组、20%白酒+单纯性肥胖模型组、40%白酒+单纯性肥胖模型组、60%白酒+单纯性肥胖模型组,每组6只。用高脂饲料喂养建立单纯性肥胖大鼠模型,并以不同浓度的白酒和生理盐水分别灌胃饲养1w,随后对各组大鼠心脏取血,制作血清样本,检测血清中谷丙转氨酶(Alanine aminotransferase,ALT)、谷草转氨酶(Aspartate transaminase,AST)含量,并取其肝脏制作HE染色切片。结果:病理切片显示20%、40%、60%白酒+单纯性肥胖模型组肝小叶周边细胞出现不同程度水肿、脂肪变、坏死。生化分析表明20%、40%、60%白酒+单纯性肥胖模型组DeRitis比值(AST/ALT)较空白对照组和单纯性肥胖模型组均有增高,且此三组的DeRisit比值随白酒浓度升高而增加。结论:20%、40%、60%白酒+单纯性肥胖模型组大鼠肝细胞有不同程度病理损伤。饮酒对单纯性肥胖大鼠肝脏的损伤具有剂量依赖性。
Objective:To study the effects of different concentrations of alcohol on liver enzymes and liver structure on nutritional obesity rats.Methods:Thirty SD rats were randomly divided into five groups:nutritional obesity rat model group,nutritional obesity rat model group with 20% alcohol intake,nutritional obesity rat model group with 40% alcohol intake,nutritional obesity rat model group with 60% alcohol intake,control group.The nutritional obesity rat model was established by feeding high-fat diet.Intragastric administration of different concentrations of alcohol and saline were performed for each group for 1week,then detecting AST and ALT value viacardiac blood sample,and liver histological slices were analysed.Results:From the pathological sections of nutritional obesity rat model group with 20%,40%,60% alcohol intake,it can be seen that varying degrees of celluar steatosis,edema,inflammation and necrosis of around the hepatic lobule occured.Biochemical analysis showed that the DeRitis ratios(AST/ALT)of nutritional obesity rat model group with 20%,40%,60% alcohol intake were significantly higher than that of nutritional obesity rat model group and normal control group,the DeRitis ratios(AST/ALT)of three groups increased with the increasing of the concentration of alcohol.Conclusion:Liver cells of rats in three models were damaged to different extent.The liver injury in rats drinking on nutritional obesity was dose dependent.
